The yield of levan extracted from microbial fermentation broth is low, so in vitro catalytic synthesis by levansucrase expected to be one the industrial production approaches levan. recombinant plasmid pET-28a-AcmA-Z constructed a previous study was used produce levansucrase. effects temperature, pH, and metal ions on formation activity were investigated. polymer analyzed means HPIC, FTIR, NMR techniques. could easily purified step enzyme had single band clearly visible SDS-PAGE. conditions for enzymatic reactions optimal at pH 5.2 40 ℃, enzymes stimulated K+ Ca2+. biosynthesis 10% (w/v) sucrose with 6.45 U/g 30.6 g/L. molecular weight approximately 1.56 × 106 Da, as measured GPC. HPIC analysis showed that monosaccharide composition fructose glucose. results FTIR indicated produced β-(2,6) this provide basis large-scale methods.
