چکیده
OssiMend® Bioactive (Collagen Matrix Inc., NJ) is a three-component porous composite bone graft device of 45S5 Bioglass/carbonate apatite/collagen. Our in vitro studies showed that conditioned media the dissolution products OssiMend stimulated primary human osteoblasts to form mineralized bone-like nodules one week, basal culture (no osteogenic supplements). Osteoblast differentiation was followed by gene expression analysis and mineralization assay. In contrast, from commercial (Bioglass-free carbonate apatite/collagen scaffolds), or Bioglass particulate alone, did not induce extracellular matrix, but osteoblast mature osteoblasts, evidenced strong upregulation BGLAP IBSP mRNA levels. The calcium ions soluble silicon species released particles additional phosphorus release mediated osteostimulatory effects. Medium with had much higher concentration than alone. While led precipitation cell media, produced OssiMend. These results suggest adding produces synergistic osteostimulation effect on osteoblasts. summary, scaffold (OssiMend® Bioactive) stimulate matrix without any supplements. faster for ordinary Bioglass.

Culture & differentiation of mesenchymal stem cell into osteoblast on degradable biomedical composite scaffold: In vitro study
BACKGROUND & OBJECTIVES There is a significant bone tissue loss in patients from diseases and traumatic injury. The current autograft transplantation gold standard treatment has drawbacks, namely donor site morbidity and limited supply. The field of tissue engineering has emerged with a goal to provide alternative sources for transplantations to bridge this gap between the need and lack of bone...
متن کاملOsteoblast Differentiation on Collagen Scaffold with Immobilized Alkaline Phosphatase
Background In tissue engineering, scaffold characteristics play an important role in the biological interactions between cells and the scaffold. Cell adhesion, proliferation, and activation depend on material properties used for the fabrication of scaffolds. Objective In the present investigation, we used collagen with proper characteristics including mechanically stability, biodegradability ...

Binding of integrin adhesion receptors to extracellular matrix components, such as fibronectin and type I collagen, activates signaling pathways directing osteoblast survival, cell-cycle progression, gene expression, and matrix mineralization. Biomimetic strategies exploit these adhesive interactions to engineer bio-inspired surfaces that promote osteoblast adhesion and differentiation, bone fo...
متن کاملThe Effects of Iron Oxide Nanoparticle on Differentiation of Human Mesenchymal Stem Cells to Osteoblast
Introduction: IIron oxide nanoparticles (IO NP) have an increasing number of biomedical applications. To date, the potential cytotoxicity of these particles remains an issue of debate. Little is known about the cellular interaction or toxic effects of IO NP on differentiation of stem cells. The aim of the present study was to investigate the possible toxic role of different doses of IO NP in di...

The aim of this study was to develop a three-dimensional porous bone graft material as vehicle for simvastatin delivery and to investigate its effect on primary human osteoblasts from three donors. Highly porous titanium dioxide (TiO2) scaffolds were submerged into simvastatin containing alginate solution. Microstructure of scaffolds, visualized by scanning electron microscopy and micro-compute...
