Biodelivery of therapeutic extracellular vesicles: should mononuclear phagocytes always be feared?

چکیده
At present, extracellular vesicles (EVs) are considered key candidates for cell-free therapies, including treatment of allergic and autoimmune diseases. However, their therapeutic effectiveness, dependent on proper targeting to the desired cells, is significantly limited due reduced bioavailability resulting from rapid clearance by cells mononuclear phagocyte system (MPS). Thus, developing strategies avoid EV elimination essential when applying them in clinical practice. On other hand, malfunctioning MPS contributes various immune-related pathologies. Therapeutic reversal these effects with EVs would be beneficial could achieved, example, modulating macrophage phenotype or regulating antigen presentation dendritic cells. Additionally, intended macrophages replication repackaging molecules into new vesicle subtype can allow specific appropriate populations acceptor Herein, we briefly discuss under-explored aspects MPS-EV interactions that undoubtedly require further research order accelerate use EVs.

EXTRACELLULAR VESICLES AND THEIR THERAPEUTIC APPLICATIONS: A REVIEW
Background & Aims: Extracellular vesicles are double phospholipid layer vesicles releasing from several cells. These particles contain bio-molecules including mRNA, miRNA, DNA, proteins, and lipids which play a pivotal role in the cell to cell communication. Once delivered, they participate to change target cell function, fate, morphology, differentiation, and growth. The present study is a re...
متن کاملTherapeutic Potential of Extracellular Vesicles
Extracellular vesicles (EV) have emerged as important mediators of intercellular communication. By their origin, we can find vesicles derived from plasma membrane such as microvesicles, ectosomes, and membrane particles or exosomes, which originate in the endosomal membrane compartment. They contain numerous proteins, lipids, and even nucleic acids like mRNA and miRNA that can affect the cells ...
متن کاملFocus on Extracellular Vesicles: Therapeutic Potential of Stem Cell-Derived Extracellular Vesicles
The intense research focus on stem and progenitor cells could be attributed to their differentiation potential to generate new cells to replace diseased or lost cells in many highly intractable degenerative diseases, such as Alzheimer disease, multiple sclerosis, and heart diseases. However, experimental and clinical studies have increasingly attributed the therapeutic efficacy of these cells t...
متن کاملThe Differentiation of Mononuclear Phagocytes
The in vitro differentiation of homogeneous populations of monocyte-like cells from the unstimulated mouse peritoneal cavity is described. Under the conditions employed, a progressive increase in cell size occurs without significant cell division. This process is characterized morphologically by the accumulation of phase-dense and neutral red-positive granules, mitochondria, and lipid droplets....
متن کاملFunctional Polarization of Mononuclear Phagocytes
Functional Polarization of Mononuclear Phagocytes Mononuclear phagocyte (MP) encompass bone marrow precursor cells and peripheral blood monocytes that, after a very short time (1–2 days), migrate into the different organs and tissues. Under the influence of the different microenvironments, recently migrated monocytes give rise to a variety of MP subtypes, including mucosal macrophages, dendriti...
