Bacterial Outer Membrane Vesicles as Nano?Scale Bioreactors: A Fatty Acid Conversion Case Study

نویسندگان

چکیده

Bacterial outer membrane vesicles (OMVs) are small unilamellar proteoliposomes, involved in various functions including cell-to-cell signalling and protein excretion. We have engineered the OMVs of Escherichia coli to nano-scaled bioreactors for biotransformation fatty acids by targeting a acid double bond hydratase Stentrophomonas maltophilia (SmOhyA) and/or photoactivated decarboxylase from Chlorella variabilis NC64 A (CvFAP) into OMVs. Engineered containing both SmOhyA CvFAP were able catalyse transformation oleic ((Z)-octadec-9-enoic acid) 9-hydroxyheptadecane via (R)-10-hydroxyoctadecanoic acid. The specific rates reached 8.0×10?12 ?mol/min per OMV.

Bacterial Outer Membrane Vesicles and Vaccine Applications

Vaccines based on outer membrane vesicles (OMV) were developed more than 20 years ago against Neisseria meningitidis serogroup B. These nano-sized structures exhibit remarkable potential for immunomodulation of immune responses and delivery of meningococcal antigens or unrelated antigens incorporated into the vesicle structure. This paper reviews different applications in OMV Research and Devel...

Glycoengineered Outer Membrane Vesicles: A Novel Platform for Bacterial Vaccines.

The World Health Organization has indicated that we are entering into a post-antibiotic era in which infections that were routinely and successfully treated with antibiotics can now be lethal due to the global dissemination of multidrug resistant strains. Conjugate vaccines are an effective way to create a long-lasting immune response against bacteria. However, these vaccines present many drawb...

Outer membrane vesicles as platform vaccine technology

Outer membrane vesicles (OMVs) are released spontaneously during growth by many Gram-negative bacteria. They present a range of surface antigens in a native conformation and have natural properties like immunogenicity, self-adjuvation and uptake by immune cells which make them attractive for application as vaccines against pathogenic bacteria. In particular with Neisseria meningitidis, they hav...

Bacterial outer membrane vesicles at the plant–pathogen interface

Gram-negative bacteria outer membrane vesicles (OMVs) are extracellularly released blebs, constantly detaching from the bacterial cell surface. Being ubiquitous among bacteria and diverse in content, OMVs have a plethora of functions: promoting virulence, mediating bacterial cell–cell communication, modulating host immune response, and more. Though most research on OMVs has been carried out on ...

Bacterial outer membrane vesicles and the host-pathogen interaction.

Extracellular secretion of products is the major mechanism by which Gram-negative pathogens communicate with and intoxicate host cells. Vesicles released from the envelope of growing bacteria serve as secretory vehicles for proteins and lipids of Gram-negative bacteria. Vesicle production occurs in infected tissues and is influenced by environmental factors. Vesicles play roles in establishing ...
