چکیده
Aberrant proliferation and migration of residual lens epithelial cells (LECs) is the main cause posterior capsule opacification (PCO) after cataract surgery. The purpose this study was to confirm protective effects Astragaloside IV (AST) in transforming growth factor beta (TGF-β)-induced LECs. Human LECs, SRA01/04, were used study. MTT assay EdU perform cell viability proliferation, respectively. cycle determined using flow cytometry. Cell invasion transwell wound healing assay, Protein expression examined western blotting. Type I collagen (Col IV) fibronectin (FN) also detected immunofluorescence (IF). viability, suppressed by AST a concentration-dependent manner TGF-β-treated stopped at G2 phase manner. Expression Cyclin B1 CDK1 FN, Col I, IV, α-SMA (α-smooth muscle actin), Snail Slug downregulated inhibited protein Jagged-1, Notch1, Notch2, Notch3, Hes-1 Hey-1. Taken together, data from demonstrated that LEC invasion, as well fibroblastic differentiation epithelial-mesenchymal transition process through inhibition Jagged-1/Notch pathway, providing potential supplemental treatment for PCO.

Effect of Transforming Growth Factor-β (TGF-β) on proliferation of gastric epithelial cells in culture
Objective: Helicobacter pylori has a well-established role in the development of gastric cancer. In vitro studies reveal increased proliferation of the gastric mucosa in the presence of H. pylori infection. It has been also shown that production of some cytokines, such as interleukin-1 beta (IL-1b) is in...
متن کاملTanshinone IIA inhibits AGEs-induced proliferation and migration of cultured vascular smooth muscle cells by suppressing ERK1/2 MAPK signaling
Objective(s): Vascular smooth muscle cells (VSMCs) play a key role in the pathogenesis of diabetic vascular disease. Our current study sought to explore the effects of tanshinone IIA on the proliferation and migration of VSMCs induced by advanced glycation end products (AGEs). Materials and Methods: In this study, we examined the effects of tanshinone IIA by cell proliferation assay and cell mi...
متن کاملBortezomib inhibits proliferation, migration, and TGF-β1–induced epithelial–mesenchymal transition of RPE cells
Purpose Nuclear factor kappa B (NF-κB) plays an important role in the epithelial-mesenchymal transition (EMT) of RPE cells. We investigated the effects of a proteasome inhibitor, bortezomib, on the EMT in RPE cells. In addition, we assessed the influence of bortezomib on regulation of the NF-κB pathway during this process. Methods After treatment with various concentrations of bortezomib, cel...
متن کاملShikonin inhibits the proliferation of human lens epithelial cells by inducing apoptosis through ROS and caspase-dependent pathway.
Shikonin is a compound from the herbal plant Lithospermum erythrorhizon that has been proved to possess powerful anti-proliferative effect on many kinds of cancers and to be safe in in vivo study. Posterior capsular opacification (PCO), the most frequent complication of cataract surgery, is mainly caused by the uncontrolled proliferation of retained human lens epithelial cells (HLEs). In this s...
