Assessment of Hyperosmolar Blood–Brain Barrier Opening in Glioblastoma via Histology with Evans Blue and DCE-MRI

Background: While the blood–brain barrier (BBB) is often compromised in glioblastoma (GB), perfusion and consequent delivery of drugs are highly heterogeneous. Moreover, accessibility largely impaired margins tumor for infiltrating cells at origin recurrence. In this work, we evaluate value methods to assess hemodynamic changes induced by a hyperosmolar shock core GB model. Methods: Osmotic was with an intracarotid infusion hypertonic solution mannitol mice grafted U87-MG cells. The distribution fluorescent dye (Evans blue) within brain assessed via histology. Dynamic contrast-enhanced (DCE)-MRI injection Gadolinium-DOTA as contrast agent also used effect on parameters diffusion outside area. Results: histological study revealed that diffused much more area after osmotic than control tumors. However, DCE-MRI did not reveal any change permeability BBB, whatever studied MRI parameter. Conclusions: use seems be promising method increase compounds GB. Nevertheless, analysis using gadolinium-DOTA limited determining efficacy opening BBB shock.
