Assembly strategy of liposome and polymer systems for siRNA delivery

In recent years, gene therapy has made tremendous progress in the development of disease treatment. Among them, siRNA offers specificity silencing, ease synthesis, and short period, been intensively studied worldwide. However, as hydrophilic polyanion is easily degraded vivo poorly taken up into cells so, benefits its powerful silencing ability will not be realized until better carriers are developed that capable protecting delivering it intact to cytoplasm target cells. Cationic liposomes (CL) cationic polymers (CP) main non-viral vectors, there have a lot reports on use these two deliver siRNA. Whereas, far we know, few review articles provide an in-depth summary loading principle internal structures delivery system. We summarize formation assembly structure liposome-siRNA polymer-siRNA complexes, point out their advantages characteristics also show how perfect improve clinical application future. It supports some useful suggestions for therapy, specifically, safe efficient delivery.
