Applications of CRISPR-Cas9 Technology to Treat CCR5 Tropic HIV-1

چکیده
Human immunodeficiency virus type 1 (HIV-1) is most notable for its role in directly infecting and killing CD4 T cells the immune system, which can lead to acquisition of Acquired Immunodeficiency Syndrome (AIDS). Currently, there are no cures HIV-1, leaving 37 million people with HIV-1 infections definitive treatment. Researchers working towards developing long-term treatments using CRISPR-Cas9. These therapies include genetically engineering antiviral resistance into vulnerable cell types cutting out viral genomes from infected cells. This article will explain life cycle detail, give an overview CRISPR-Cas9 technology, then go detail about potential applications treatment CCR5 Tropic HIV-1.

Application of CRISPR/Cas9 Technology to HBV.
More than 240 million people around the world are chronically infected with hepatitis B virus (HBV). Nucleos(t)ide analogs and interferon are the only two families of drugs to treat HBV currently. However, none of these anti-virals directly target the stable nuclear covalently closed circular DNA (cccDNA), which acts as a transcription template for viral mRNA and pre-genomic RNA synthesis and s...
متن کاملProgress in HIV-Carbohydrate Interactions: Infectivity of CCR5-Tropic HIV-1 is Enhanced by GP120 from CXCR4-Tropic Virus
Human immunodeficiency virus type-1 is highly variable and utilizes two independent pathways, one via the CCR5 coreceptor and the other via CXCR4 to enter CD4+ cells. Both virus types can simultaneously be found in HIV-1-infected patients and the shift to a majority of CXCR4-tropic viruses is an indication for the development of AIDS-related symptoms. It is thought that each virus type will inf...

Targeted mutagenesis based on homologous recombination has been a powerful tool for understanding the mechanisms underlying development, normal physiology, and disease. A recent breakthrough in genome engineering technology based on the class of RNA-guided endonucleases, such as clustered regularly interspaced short palindromic repeats (CRISPR)-associated Cas9, is further revolutionizing biolog...
متن کاملGrowth potentials of CCR5-tropic/CXCR4-tropic HIV-1mt clones in macaque cells
Human immunodeficiency virus type 1 (HIV-1) is strictly adapted to humans, and cause AIDS only in humans. Consequently, no experimental animals susceptible to HIV-1 and suitable for the AIDS model study are available to date (Nomaguchi et al., 2008, 2011). To overcome this issue, viruses genetically related to HIV-1 have been challenged into macaque monkeys to mimic the natural HIV-1 infection....

Recently several gene-editing technologies developed are being explored for their potential utility in providing new and unique treatments for HIV. One of these technologies is the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated (Cas)9 system. This system is being explored for its utility against host genes important to HIV infection, namely the HIV corecept...
