Antioxidative stress effects of Humulus japonicus extracts on neuronal PC12 cells

The objective of the current study was to assess antioxidant activity different solvents containing Humulus japonicus extract (HJE) and their effects on neurons. H. ethanol extracts (HEEs) together with water (HWEs) were generated via extraction leaves 30%, 50%, 80% solutions under a hot temperature 121 °C. HEEs showed higher levels bioactive compounds, such as total flavonoids polyphenols, stronger activities in contrast HWEs. HEE (80%) highest content compounds activity. Next, we used H2O2-induced rat model pheochromocytoma PC12 cells investigate cellular signal regulation HEEs. results that HJEs resisted oxidative stress MAPK and/or AKT-NRF2 NF-kappa B signaling pathways upregulated enzymes SOD, catalase, HO-1, OGG1, thereby reducing intracellular ROS levels. Further, HJE promoted neuronal survival by upregulating sirtuin 1 expression. This provides evidence supporting potential benefits preventing cells, suggesting its application promising functional food additive or therapeutic agent for prevention treatment stress-related neurodegenerative diseases.
