Systemic Lupus Erythematosus (SLE) is an autoimmune inflammatory condition with a wide spectrum of disease manifestations and severities, resulting in significant morbidity mortality. The aetiopathogenesis SLE complex. Young women certain ethnicities are commonly affected, suggesting hormonal genetic influence. Diverse immunological abnormalities have been described. A characteristic abnormality the presence autoantibodies, implicating central role for B cells pathogenesis and/or perpetuation. Whilst conventional therapies improved outcomes, great unmet need remains. Recently, biological being explored. B-cell depletion therapy rituximab has use off-label nearly two decades. Inconsistent results between uncontrolled controlled studies raised doubts about its efficacy. In this review, we will focus on cell rationale behind anti-CD20 monoclonal antibody (mAb), rituximab, be explored including evaluation clinical trial experience. Finally, discuss mechanistic basis considering alternative mAbs.
