چکیده
Background: Nanotechnology symbolizes a broad discipline with enormous potential in cancer treatment bridging one of the bottlenecks traditional approaches therapy which is an inability to deliver adequate quantities anti-cancer drugs tumor area. Studies on nanoparticles indicate their importance angiogenesis and metastasis. Aim: The present study assessed anti-angiogenesis anti-metastatic effects biogenic silver (AgNPs) synthesized from neem plant (Azadirachta indica). Methods: Chicken chorioallantoic membrane (CAM) two-dimensional (2D) wound healing assays were used anti-angiogenic AgNPs respectively. Twenty-four fertilized eggs divided into four groups: two treatments at 100 μg/ml 200 μg/ml; negative control (1% DMSO) positive (cyclophosphamide). On day 8 incubation, filter discs impregnated different concentration levels placed CAM. 12 CAMs imaged using stereomicroscope, scaled ImageJ, morphometric spatial parameters computed AngioTool software. Vessel area, vessel percent total number junctions, length, average mean lacunarity, junction density measured. crown-rump length (CRL) fetal weight also recorded 16 incubation. In order determine relative gene expression profiles iNOS VEGF, RNA was extracted CAM, qRT-PCR performed β-Actin as reference gene. For 2D assay, DU145 human prostate cells grown Dulbecco’s Modified Eagle’s Medium supplemented 10% Fetal Bovine Serum. Results: Biogenic demonstrated dose-dependent manner generated CAM images. Also, revealed down-regulation VEGF genes. 2-dimensional assay showed inhibition migration motility for 72-hours assessment. Conclusion: postulates that can prevent by inactivation VEGF-NO VEGF/VEGF-R pathways while inhibiting cell
