Analyzing Oxidative Stress in Murine Intestinal Organoids using Reactive Oxygen Species-Sensitive Fluorogenic Probe

چکیده
Reactive oxygen species (ROS) play essential roles in intestinal homeostasis. ROS are natural by-products of cell metabolism. They produced response to infection or injury at the mucosal level as they involved antimicrobial responses and wound healing. also critical secondary messengers, regulating several pathways, including growth differentiation. On other hand, excessive levels lead oxidative stress, which can be deleterious for cells favor diseases like chronic inflammation cancer. This work provides a straightforward method detect murine organoids by live imaging flow cytometry, using commercially available fluorogenic probe. Here protocol describes assaying effect compounds that modulate redox balance specific types, exemplified here analysis stem genetically labeled with GFP. may used fluorescent probes.

Oxidative stress and reactive oxygen species.
This article discusses different aspects concerning classification/nomenclature, biochemical properties and pathophysiological roles of reactive oxygen species (ROS) which are pivotal to interpret the concept of oxidative stress. In vitro studies in both the prokaryotes and eukaryotes clearly demonstrate that exogenous or constitutive and inducible endogenous sources of ROS together with cofact...
متن کاملReactive oxygen species, oxidative stress, glaucoma and hyperbaric oxygen therapy
This review examines the role of oxidative stress in damage to cells of the trabecular meshwork and associated impaired aqueous drainage as well as damage to retinal ganglion cells and associated visual field losses. Consideration is given to the interaction between vascular and mechanical explanations for pathological changes in glaucoma. For example, elevated intraocular pressure (IOP) forces...
متن کاملReactive oxygen species: metabolism, oxidative stress, and signal transduction.
Several reactive oxygen species (ROS) are continuously produced in plants as byproducts of aerobic metabolism. Depending on the nature of the ROS species, some are highly toxic and rapidly detoxified by various cellular enzymatic and nonenzymatic mechanisms. Whereas plants are surfeited with mechanisms to combat increased ROS levels during abiotic stress conditions, in other circumstances plant...
متن کاملSpectroscopic detection of reactive oxygen species and oxidative stress
Reactive oxygen species (ROS) include both free radicals and non-radical compounds. ESR spectroscopy, especially employing spin traps, is the unique technique allowing for detection, identification and quantification of free radicals in vitro and their imaging in vivo. However, the sensitivity of this technique is limited and essays based on the use of more sensitive fluorogenic and luminogenic...
متن کاملUpdate on Mitochondrial Reactive Oxygen Species Mitochondrial Reactive Oxygen Species. Contribution to Oxidative Stress and Interorganellar Signaling
The inner membrane of a plant mitochondrion contains the mitochondrial electron transport chain (mtETC), consisting of protein complexes that use an energy source-derived reductant to form a proton gradient across the membrane. This proton gradient drives ATP synthesis, a primary mitochondrial function. Ultimately, electrons in the mtETC are transferred through one of two pathways from ubiquino...
