Analysis and Characterization of Chitinase in Bacillus salmalaya Strain 139SI

Introduction: Chitinases are enzymes that hydrolyze the internal β-1,4 glycosidic linkages of chitin, a significant structural component arthropod exoskeletons and fungus cell walls. Objective: Main objective current study was to determine protein check antimicrobial, antifungal activity chitinase in Bacillus salmalaya strain 139SI. Methodology: Purified estimation were quantified by method Lowry method. Antimicrobial action done using standard Disc Diffusion while hyphal extension inhibition assay used test pure strains 139SI, 140SI, 141SI. Results: In this study, 139SIexhibited strong hemolytic their concentration measured as 56.43 mg/mL. addition, 139SI had against phytopathogenic including Fusarium sp., R. solani Phytophthora sp. Strain capability degrading peptidoglycan walls gram-negative bacteria such Escherichia coli but not gram-positive, Staphylococcus aureus. Chitinase observed when 200μl crude extract able degrade 0.09 g chitin shrimp shell breaking down structure bonds effectively early 2 days or up 7 days. Conclusion: Hence, based on results, B. has potential be novel biofunctional could use biological agent fungal shells waste many kind insect crustaceans for solving future problem agricultural sector fishery industry.
