Amyloid-Mediated Mechanisms of Membrane Disruption

چکیده
Protein aggregation and amyloid formation are pathogenic events underlying the development of an increasingly large number human diseases named “proteinopathies”. Abnormal accumulation in affected tissues ? (A?) peptide, islet polypeptide (IAPP), prion protein, to mention a few, involved occurrence Alzheimer’s (AD), type 2 diabetes mellitus (T2DM) diseases, respectively. Many reports suggest that toxic properties aggregates correlated with their ability damage cell membranes. However, molecular mechanisms causing amyloid/membrane interactions still far be completely elucidated. This review aims at describing mutual relationships linking abnormal protein conformational transition self-assembly into membrane damage. A cross-correlated analysis all these closely intertwined factors is thought provide valuable insights for comprehensive description and, turn, design effective therapies.

Ganglioside G(M1)-mediated amyloid-beta fibrillogenesis and membrane disruption.
There is increasing evidence that a class of cell membrane glycolipids, gangliosides, can mediate the fibrillogenesis and toxicity of Alzheimer's disease amyloid-beta peptide (Abeta). Using lipid monolayers and vesicles as model membranes, we measured the insertion of Abeta into 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC)-ganglioside GM1 monolayers to probe Abeta-GM1 interactions, imaged...
متن کاملDirect three-dimensional visualization of membrane disruption by amyloid fibrils.
Protein misfolding and aggregation cause serious degenerative conditions such as Alzheimer's, Parkinson, and prion diseases. Damage to membranes is thought to be one of the mechanisms underlying cellular toxicity of a range of amyloid assemblies. Previous studies have indicated that amyloid fibrils can cause membrane leakage and elicit cellular damage, and these effects are enhanced by fragment...
متن کاملCationic Peptide Exposure Enhances Pulsed-Electric-Field-Mediated Membrane Disruption
BACKGROUND The use of pulsed electric fields (PEFs) to irreversibly electroporate cells is a promising approach for destroying undesirable cells. This approach may gain enhanced applicability if the intensity of the PEF required to electrically disrupt cell membranes can be reduced via exposure to a molecular deliverable. This will be particularly impactful if that reduced PEF minimally influen...
متن کاملCalcium dysregulation and membrane disruption as a ubiquitous neurotoxic mechanism of soluble amyloid oligomers.
Increasing evidence suggests that amyloid peptides associated with a variety of degenerative diseases induce neurotoxicity in their intermediate oligomeric state, rather than as monomers or fibrils. To test this hypothesis and investigate the possible involvement of Ca2+ signaling disruptions in amyloid-induced cytotoxicity, we made homogeneous preparations of disease-related amyloids (Abeta, p...
متن کاملO-8: Molecular Mechanisms of Membrane Fusion Involved in Fertilization
Background: Assisted fertilization procedures are a currently widespread practice to regulate reproduction in humans and animals. The arising question is why the human being manipulating gametes to generate new individuals, if we do not understand yet the molecular mechanism of fertilization?. Successful completion of fertilization in mammals is dependent on three membrane fusion events: 1. the...
