Amyloid β1–42 Oligomers Induce Galectin–1S8 O–GlcNAcylation Leading to Microglia Migration

چکیده
Protein O–GlcNAcylation has been associated with neurodegenerative diseases such as Alzheimer’s disease (AD). The of the Amyloid Precursor (APP) regulates both trafficking and processing APP through amyloidogenic pathway, resulting in release aggregation Aβ1–42 peptide. Microglia clears Aβ aggregates dead cells to maintain brain homeostasis. Here, using LC-MS/MS, we revealed that oligomers modify microglia O-GlcNAcome. We identified 55 proteins, focusing our research on Galectin-1 protein since it is a very versatile from functional point view. Combining biochemical genetic approaches, demonstrated specifically target Galectin–1S8 via OGT. In addition this, Gal–1–O–GlcNAcylated form, turn, controls human migration. Given importance migration progression AD, this study reports relationship between Serine 8–O–GlcNAcylation Galectin–1 drive microglial

Amyloid beta oligomers induce impairment of neuronal insulin receptors.
Recent studies have indicated an association between Alzheimer's disease (AD) and central nervous system (CNS) insulin resistance. However, the cellular mechanisms underlying the link between these two pathologies have not been elucidated. Here we show that signal transduction by neuronal insulin receptors (IR) is strikingly sensitive to disruption by soluble Abeta oligomers (also known as ADDL...
متن کاملAmyloid β oligomers induce interleukin-1β production in primary microglia in a cathepsin B- and reactive oxygen species-dependent manner.
Amyloid β (Aβ) peptide, a causative agent of Alzheimer's disease, forms two types of aggregates: oligomers and fibrils. These aggregates induce inflammatory responses, such as interleukin-1β (IL-1β) production by microglia, which are macrophage-like cells located in the brain. In this study, we examined the effect of the two forms of Aβ aggregates on IL-1β production in mouse primary microglia....
متن کاملSoluble amyloid-β oligomers as synaptotoxins leading to cognitive impairment in Alzheimer’s disease
Alzheimer's disease (AD) is the most common form of dementia in the elderly, and affects millions of people worldwide. As the number of AD cases continues to increase in both developed and developing countries, finding therapies that effectively halt or reverse disease progression constitutes a major research and public health challenge. Since the identification of the amyloid-β peptide (Aβ) as...
متن کاملAmyloid-β oligomers set fire to inflammasomes and induce Alzheimer's pathology
Genetic and molecular studies have confirmed the central role of amyloid-ß production and fibrillation in the pathogenesis of Alzheimer’s disease (AD). However, the pathological pathways from amyloid-ß peptide oligomerization to the major pathological hallmarks of AD, such as neurofibrillary tangles, inflammation and loss of cholinergic neurons, are largely unknown. The innate immunity defence ...
متن کاملAmyloid-β oligomers stimulate microglia through a tyrosine kinase dependent mechanism.
Alzheimer's disease (AD) has been well characterized by the presence of reactive microglia, often associated with β-amyloid (Aβ) plaque deposition. The oligomeric form of Aβ peptide (Aβ(o)) has neurotoxic effects in the presence of microglia and is suggested to potentiate proinflammatory changes in microglia in AD. Primary murine microglia cultures stimulated with Aβ(o) displayed increased prot...
