چکیده
Abstract Acetolactate synthase genes (ALS) have successfully been modified for providing resistance to ALS-inhibiting herbicides in many plant species. Based on sequence and expression analyses, we confirmed VvALS1 as the best functional ALS candidate grapevine. To develop an ALS-based herbicide selection system facilitating grape transformation, firstly evaluated responses of Vitis vinifera cv Chardonnay callus young vitro shoots Thompson Seedless several representative found a typical linear response curves some herbicides, including chlorsulfuron imazapyr belonging sulfonylurea or imidazolinone families, respectively. Secondly, created constructs containing amino acid substitutions domains which are known be critical generated transgenic plants 3 using Agrobacterium-mediated transformation meristematic bulk tissues Seedless. Finally, showed that ectopic two (P191S P191T) at N-terminal region another (W568L) C-terminal VvALS resulted high shoots. Our work highlighted potential use mutations imparting selectable marker grapevine research means fostering improvement via cisgenesis, paving way developing co-editing facilitate transgene-free gene-editing.

Amino acid residues conferring herbicide resistance in tobacco acetohydroxy acid synthase.
The enzyme AHAS (acetohydroxy acid synthase), which is involved in the biosynthesis of valine, leucine and isoleucine, is the target of several classes of herbicides. A model of tobacco AHAS was generated based on the X-ray structure of yeast AHAS. Well conserved residues at the herbicide-binding site were identified, and the roles of three of these residues (Phe-205, Val-570 and Phe-577) were ...

Two-dimensional gel electrophoresis coupled to mass spectrometry analysis was used to examine for the first time the effect of a herbicide (flumioxazin) on a crop species (Vitis vinifera L.) at the proteome level. Examination of 2-D maps derived from chemically stressed tissues revealed the presence of 33 spots displaying a differential expression pattern. The presence of stress responsive prot...
متن کاملTwo consecutive aspartic acid residues conferring herbicide resistance in tobacco acetohydroxy acid synthase.
Acetohydroxy acid synthase (AHAS) catalyzes the first common step in the biosynthesis pathway of the branch chain amino acids in plants and microorganisms. A great deal of interest has been focused on AHAS since it was identified as the target of several classes of potent herbicides. In an effort to produce a mutant usable in the development of an herbicide-resistant transgenic plant, two conse...

Grapevine chloroplast (cp) DNA diversity was analysed for the first time through amplification and digestion of fragments of the large single copy (LSC) region by polymerase chain reaction-restriction fragment length polymorphism methodology and also by amplification of three microsatellite loci, previously described as polymorphic in grapevine. Thirty-eight grapevine cultivars collected mainly...
متن کاملSugars and flowering in the grapevine (Vitis vinifera L.).
Sugars play an important role in grapevine flowering. This complex process from inflorescence initiation to fruit maturity takes two growing seasons. Currently, most of the available data concern the involvement of sugars as energy sources during the formation of reproductive structures from initiation of inflorescences during the summer of the first year, until flower opening during the follow...
