Altered glucocorticoid reactivity and behavioral phenotype in rx3-/- larval zebrafish

چکیده

Introduction The transcription factor rx3 is important for the formation of pituitary and parts hypothalamus. Mutant animals lacking function have been well characterized in developmental studies, but relatively little known about their behavioral phenotypes. Methods We used cell type staining to reveal differences stress axis architecture, performed cortisol measurements behavior analysis study both hormonal responses mutants. Results Discussion Consistent with role hypothalamus development, we show a distinct loss corticotrope cells involved regulation, severe reduction innervation by hypothalamic cells, lack stress-induced release Interestingly, despite these deficits, report that rx3-/- larval zebrafish can still display nominal stressful non-stressful stimuli. However, unlike wildtypes, mutants proper pituitary-interrenal do not enhanced performance under moderate level, supporting view corticotroph are required some types stimuli modulate subtle adjustments stress.

Optogenetic elevation of endogenous glucocorticoid level in larval zebrafish

The stress response is a suite of physiological and behavioral processes that help to maintain or reestablish homeostasis. Central to the stress response is the hypothalamic-pituitary-adrenal (HPA) axis, as it releases crucial hormones in response to stress. Glucocorticoids (GCs) are the final effector hormones of the HPA axis, and exert a variety of actions under both basal and stress conditio...

Behavioral genetics in larval zebrafish: learning from the young.

Deciphering the genetic code that determines how the vertebrate nervous system assembles into neural circuits that ultimately control behavior is a fascinating and challenging question in modern neurobiology. Because of the complexity of this problem, successful strategies require a simple yet focused experimental approach without limiting the scope of the discovery. Unbiased, large-scale forwa...

An altered neonatal behavioral phenotype in Mecp2 mutant mice.

We examined somatic growth, somatosensory reflexes, and ultrasonic calls from postnatal day 3 to day 18 in Mecp2 mutant mice, a mouse model of Rett syndrome. Both Mecp2 null male and Mecp2 heterozygous female mice exhibited normal somatic growth, but transient delays in the development of some reflexes relative to sex-matched wild-type mice. Both Mecp2 null male and heterozygous female mice exh...

Sensorimotor gating in larval zebrafish.

Control of behavior in the natural environment where sensory stimuli are abundant requires superfluous information to be ignored. In part, this is achieved through selective transmission, or gating of signals to motor systems. A quantitative and clinically important measure of sensorimotor gating is prepulse inhibition (PPI) of the startle response, impairments in which have been demonstrated i...

Rx3 and Shh direct anisotropic growth and specification in the zebrafish tuberal/anterior hypothalamus

In the developing brain, growth and differentiation are intimately linked. Here, we show that in the zebrafish embryo, the homeodomain transcription factor Rx3 coordinates these processes to build the tuberal/anterior hypothalamus. Analysis of rx3 chk mutant/rx3 morphant fish and EdU pulse-chase studies reveal that rx3 is required to select tuberal/anterior hypothalamic progenitors and to orche...
