AHNAK controls 53BP1-mediated p53 response by restraining 53BP1 oligomerization and phase separation

چکیده

Summary p53-binding protein 1 (53BP1) regulates both the DNA damage response and p53 signaling. Although 53BP1's function is well established in double-strand break repair, how its role signaling modulated remains poorly understood. Here, we identify scaffolding AHNAK as a G1 phase-enriched interactor of 53BP1. We demonstrate that binds to 53BP1 oligomerization domain controls multimerization potential. Loss results hyper-accumulation on chromatin enhanced phase separation, culminating an elevated response, compromising cell survival cancer cells but leading senescence non-transformed cells. Cancer transcriptome analyses indicate AHNAK-53BP1 cooperation contributes suppression target gene networks tumors loss sensitizes combinatorial treatments. These findings highlight rheostat function, which surveys proliferation by preventing excessive response.

53BP1 promotes microhomology-mediated end-joining in G1-phase cells

Alternative non-homologous end joining (alt-NHEJ) was originally identified as a backup repair mechanism in the absence of classical NHEJ (c-NHEJ) factors but recent studies have demonstrated that alt-NHEJ is active even when c-NHEJ as well as homologous recombination is available. The functions of 53BP1 in NHEJ processes are not well understood. Here, we report that 53BP1 promotes DNA double-s...

Role of 53BP1 oligomerization in regulating double-strand break repair.

Tumor suppressor p53-binding protein 1 (53BP1) regulates the repair of dysfunctional telomeres lacking the shelterin protein TRF2 by promoting their mobility, their nonhomologous end-joining (NHEJ), and, as we show here, by blocking 5' resection by CtIP. We report that these functions of 53BP1 required its N-terminal ATM/ATR target sites and its association with H4K20diMe, but not the BRCT doma...

53BP1 and p53 synergize to suppress genomic instability and lymphomagenesis.

p53-binding protein 1 (53BP1) participates in the cellular response to DNA double-stranded breaks where it associates with various DNA repair/cell cycle factors including the H2AX histone variant. Mice deficient for 53BP1 (53BP1(-/-)) are sensitive to ionizing radiation and immunodeficient because of impaired Ig heavy chain class switch recombination. Here we show that, as compared with p53(-/-...

RNF168 ubiquitylates 53BP1 and controls its response to DNA double-strand breaks.

Defective signaling or repair of DNA double-strand breaks has been associated with developmental defects and human diseases. The E3 ligase RING finger 168 (RNF168), mutated in the human radiosensitivity, immunodeficiency, dysmorphic features, and learning difficulties syndrome, was shown to ubiquitylate H2A-type histones, and this ubiquitylation was proposed to facilitate the recruitment of p53...

Functional interaction between BLM helicase and 53BP1 in a Chk1-mediated pathway during S-phase arrest

Bloom's syndrome is a rare autosomal recessive genetic disorder characterized by chromosomal aberrations, genetic instability, and cancer predisposition, all of which may be the result of abnormal signal transduction during DNA damage recognition. Here, we show that BLM is an intermediate responder to stalled DNA replication forks. BLM colocalized and physically interacted with the DNA damage r...
