Aging-associated prostate smooth muscle hypercontractility in rats

Benign prostatic hyperplasia (BPH) is a multifactorial disease, highly associated with aging and characterized by increased prostate smooth muscle (PSM) contractility. Animal models have been employed to explore the aging-associated PSM hypercontractility; however, studies focused in old animals, neglecting initial alterations early ages. The determination of dysfunctions onset crucial understand BPH pathophysiology propose new treatments. Considering that contractility 10-month-old rats has already explored, aim present study was characterize younger rats. Male Wistar control (3.5-month-old), 6- 8-month-old were used. Concentration-response curves phenylephrine electrical-field stimulation (EFS) conducted from all groups. For first time, we showed exhibit hypercontractility. starts around at 6-month-old, worsening during aging. exhibited hypercontractility EFS compared 6-month-old Reduced potency observed rats, indicating an age-dependent sensibility this agonist. Collectively, our findings support use aged as BPH.
