Affimer-based impedimetric biosensors for fibroblast growth factor receptor 3 (FGFR3): a novel tool for detection and surveillance of recurrent bladder cancer

نویسندگان

چکیده

Fibroblast growth factor receptor 3 (FGFR3) is a transmembrane tyrosine kinase protein in the fibroblast family, which plays key role many biological processes. Over-expression and activating mutations FGFR3 are frequent non-invasive bladder cancer, highlighting this as potential biomarker for recurrent cancer detection. Affimer reagents isolated against recombinant were assessed their affinity using double-sandwich ELISA SPR. Anti-FGFR3-14 FGFR3-21 proteins showed strong binding to used fabrication of impedimetric electrochemical biosensors. A decrease impedance was observed when sensors exposed increasing concentrations FGFR3. The successful biosensors capable detecting sub-pM nM phosphate buffered saline well synthetic urine.

The fibroblast growth factor receptor 3 (FGFR3) mutation is a strong indicator of superficial bladder cancer with low recurrence rate.

We analyzed the possible prognostic value of the recently discovered fibroblast growth factor receptor 3 (FGFR3) mutations in bladder cancer. A FGFR3 mutation was found in 34 of 53 pTaG1-2 bladder cancers, whereas none of the 19 higher-staged tumors had a mutation (P < 0.0001). In 57 patients with superficial disease followed prospectively by cystoscopy for 12 months, 14 of 23 patients in the w...

Incidence of fibroblast growth factor receptor 3 gene (FGFR3) A248C, S249C, G372C, and T375C mutations in bladder cancer.

Bladder cancer is the most frequent cancer of the urinary system. Fibroblast growth factor receptors (FGFR) belong to the tyrosine kinase family and have important roles in cell differentiation and proliferation and embryogenesis. FGFR3 is located on chromosome 4p16.3, and missense mutations of FGFR3 are associated with autosomal dominant human skeletal disorders and have some oncogenic effects...

Identification of Fibroblast Growth Factor Receptor 3 (FGFR3) as a Protein Receptor for Botulinum Neurotoxin Serotype A (BoNT/A)

Botulinum neurotoxin serotype A (BoNT/A) causes transient muscle paralysis by entering motor nerve terminals (MNTs) where it cleaves the SNARE protein Synaptosomal-associated protein 25 (SNAP25206) to yield SNAP25197. Cleavage of SNAP25 results in blockage of synaptic vesicle fusion and inhibition of the release of acetylcholine. The specific uptake of BoNT/A into pre-synaptic nerve terminals i...

Fibroblast growth factor receptor 3 is a rational therapeutic target in bladder cancer.

Activating mutations of fibroblast growth factor receptor-3 (FGFR3) have been described in approximately 75% of low-grade papillary bladder tumors. In muscle-invasive disease, FGFR3 mutations are found in 20% of tumors, but overexpression of FGFR3 is observed in about half of cases. Therefore, FGFR3 is a particularly promising target for therapy in bladder cancer. Up to now, most drugs tested f...
