Adipokines, ageing, and vascular damage

چکیده

The main physiologic function of adipokines on the energy metabolism and cardiovascular homeostasis, among others, are well known. In last years, our laboratory is providing increasing evidence about possible role as mediators vascular damage, by inducing different pro-inflammatory pro-senescent mechanisms. secretion like visfatin or dipeptidylpeptidase-4 (DPP4) adipose tissue, particularly visceral fat, enhanced in obese type 2 diabetic patients. Both enzymes metabolically active that can induce activation specific receptors, namely 4 “toll receptors” (TLR4) for “protease activated receptor 2” (PAR2) DPP4. Stimulation such receptors triggering known mechanisms, nuclear transcription factor-κB (NF-κB) inflammasome NLRP3 (“nucleotide-binding, leucine-rich-repeat, pyrin domain containing 3”), a cell structure transforms immature forms classic cytokines their derivates, interleukin-1β (IL-1β) interleukin 18 (IL-18). It worth to note these final effectors harmful effects its blockade be very relevant therapeutic approach. experiments, antagonism IL-1 with anakinra prevents inflammatory senescent evoked Moreover, this finding agreement data from other researchers, results CANTOS clinical trial, which demonstrate important cardioprotective effect mediated anti-inflammatory monoclonal antibody canakinumab. On hand, we have also provided experimental vasculopotective adipokines, angiotensin-(1-7), able induce, through Mas expression klotho protein antioxidant pathways, Nrf2-HO-1 (“nuclear factor erythroid-2 y hemoygenase-1”).

Ageing and vascular ageing.

There is an age related decline in various physiological processes. Vascular ageing is associated with changes in the mechanical and the structural properties of the vascular wall, which leads to the loss of arterial elasticity and reduced arterial compliance. Arterial compliance can be measured by different parameters like pulse wave velocity, augmentation index, and systemic arterial complian...

Androgens, ageing and vascular function.

We will never be perfect in achieving a diagnosis in every patient, and the challenge of geriatrics is how difficult it can be in some patients, and indeed, whether it is appropriate in others. An audit of our own ill-defined diagnoses showed that re-admission rates, at 1 month, were 10%, very similar to our better defined patients, and the great majority did not return to hospital within the n...

DNA Damage During Seed Ageing

Seed ageing is influenced by two environmental factors, RH and temperature. The deterioration of the stored seed is a natural phenomenon and the seeds tend to lose viability even under ideal storage conditions. Accelerated ageing has been recognized as a good predictor of the storability of seed lots. Aged seeds show decreased vigour and produce weak seedlings that are unable to survive once re...

The role of oxysterols in vascular ageing.

The ageing endothelium progressively loses its remarkable and crucial ability to maintain homeostasis of the vasculature, as it acquires a proinflammatory phenotype. Cellular and structural changes gradually accumulate in the blood vessels, and markedly in artery walls. Most changes in aged arteries are comparable to those occurring during the atherogenic process, the latter being more marked: ...

Vascular biology of ageing—Implications in hypertension

Ageing is associated with functional, structural and mechanical changes in arteries that closely resemble the vascular alterations in hypertension. Characteristic features of large and small arteries that occur with ageing and during the development of hypertension include endothelial dysfunction, vascular remodelling, inflammation, calcification and increased stiffness. Arterial changes in you...
