چکیده

Nitric oxide as a therapeutic approach to the treatment of cardiovascular diseases attracted attention researchers at end 19th century. As vasodilator, nitric may be unique agent for hypertension and, result, renal failure and left ventricular hypertrophy. The aim article is analyze literature data on possible ways modulating activity various synthases an endothelial dysfunction. Materials methods. When searching materials writing review article, such abstract databases PubMed, Google Scholar, e-Library, etc., were used. search was carried out publications period from 1990 2021. following words phrases chosen parameters selection: oxide; NO synthase; dysfunction; synthase activator; inhibitor. Results. presents history discovery its biological role, process biosynthesis, well isoforms synthesizing enzymes (NOS): neuronal – nNOS, eNOS inducible iNOS, their role in normal pathological physiology. NOS uncoupling (its molecular mechanisms) has been considered basis examples pharmacological correction (BH 4 , arginase inhibitors, statins, resveratrol) are presented. In addition, activators (calcium dobesilate, cavNOxin, some transcription activators), non-selective (L-NMMA, 1-NNA, L-NAME, ADMA, 546C88, VAS203) selective (L-NIO, 7-nitroindazole, aminoguanidine, L-NIL, GW273629, GW274150, cavtratin) inhibitors synthasehave described. Conclusion. continue promising targets development agents that modulate correct pathologies. approach, modulation can implemented treat dysfunction, which cause complications many diseases.

Arginase reciprocally regulates nitric oxide synthase activity and contributes to endothelial dysfunction in aging blood vessels.

BACKGROUND Although abnormal L-arginine NO signaling contributes to endothelial dysfunction in the aging cardiovascular system, the biochemical mechanisms remain controversial. L-arginine, the NO synthase (NOS) precursor, is also a substrate for arginase. We tested the hypotheses that arginase reciprocally regulates NOS by modulating L-arginine bioavailability and that arginase is upregulated i...

Perspective Series: Nitric Oxide and Nitric Oxide Synthases Cellular and Molecular Mechanisms of Endothelial Cell Dysfunction

The clinical relevance of the vascular L -arginine/nitric oxide (NO) 1 synthase system to vascular biology lies in the oftrepeated observation that it is altered by a variety of pathophysiological conditions. Such alterations of NO production and/or bioavailability have been shown to occur both in experimental animal models and in human subjects, in the setting of such diverse disorders as hype...

Folic acid reverts dysfunction of endothelial nitric oxide synthase.

5-methyltetrahydrofolate (MTHF), the active form of folic acid, has been reported to restore NO status in hypercholesterolemic patients. The mechanism of this effect remains to be established. We assessed the effects of L- and D-MTHF on tetrahydrobiopterin (BH(4))-free and partially BH(4)-repleted endothelial NO synthase (eNOS). Superoxide production of eNOS and the rate constants for trapping ...
