Activity Levels of Amine Transaminases Correlate with Active Site Hydrophobicity

Abstract Amine transaminases (ATAs) are biocatalysts for the synthesis of chiral amines and can be identified in sequence databases by specific motifs. This study shows that activity level towards model substrate 1‐phenylethylamine predicted solely from sequence. To demonstrate this, 15 putative ATAs with a different distribution hydrophobic or hydrophilic amino acid side chains near active site were characterized. Hydrophobic associated high better predictor than global identity to known low activities. Enzyme stability investigations revealed four out showed good operational stability.
