Activation of RANKL-producing cells under simulated microgravity with a three-dimensional clinostat in regenerating goldfish scales

چکیده
Exposure to microgravity during space flight has caused astronauts experience decreased bone density, resulting from resorption by osteoclastic activation. Activation of the nuclear factor-?B ligand (RANKL) RANKL-producing cells, a known osteoclastogenesis-promoting factor, seems be induced under microgravity. However, role cells not yet been demonstrated due lack suitable in vitro organ culture osteoblasts, osteoclasts, and osteocytes with intact matrix Previous reports that were detected specific antiserum for goldfish RANKL regenerating scales. In this study, response simulated three-dimensional clinostat was examined using system addition, mRNA expression analysis osteoblastic markers via quantitative real-time PCR conducted together histological cells. After 4 days exposure microgravity, number immune-positive increased compared control The Rankl scales after treatments significantly higher than (1G) osteoprotegerin (Opg), an osteoclastogenesis inhibitory conditions. ratio Rankl/Opg microgravity-treated 1G control. change implied activation is promoted treatments. Actually, exposed markers, such as receptor activator NF?B, cathepsin K, integrin beta-3, cellular-Src, increased. On other hand, (collagen type I alpha 1 osteocalcin) decreased. changes consistent our previously reported experiment on International Space Station. To best knowledge, we are first demonstrate conditions We strongly emphasize fish serve excellent model gravitational responses while maintaining similar vivo.

Plant growth processes in Arabidopsis under microgravity conditions simulated by a clinostat.
The life cycle of Arabidopsis plants was examined by growing them on a horizontal clinostat. Seeds on agar media were allowed to germinate and seedlings were grown under a simulated microgravity on a horizontal clinostat. Clinorotation (3 rpm) did not appear to interfere with germination of plant seeds and development of cotyledons and leaves. Stress relaxation parameters of the cell wall, the ...
متن کاملMechanisms of three-dimensional growth of thyroid cells during long-term simulated microgravity
Three-dimensional multicellular spheroids (MCS) of human cells are important in cancer research. We investigated possible mechanisms of MCS formation of thyroid cells. Both, normal Nthy-ori 3-1 thyroid cells and the poorly differentiated follicular thyroid cancer cells FTC-133 formed MCS within 7 and 14 days of culturing on a Random Positioning Machine (RPM), while a part of the cells continued...
متن کاملTime-averaged simulated microgravity (taSMG) inhibits proliferation of lymphoma cells, L-540 and HDLM-2, using a 3D clinostat
BACKGROUND Gravity is omnipresent on Earth; however, humans in space, such as astronauts at the International Space Station, experience microgravity. Long-term exposure to microgravity is considered to elicit physiological changes, such as muscle atrophy, in the human body. In addition, certain types of cancer cells demonstrate inhibited proliferation under condition of time-averaged simulated ...
متن کاملTransgenic Arabidopsis with iaaH under simulated microgravity conditions
Growth and development, and auxin polar transport in Arabidopsis thaliana transformed with iaaH gene were studied under simulated microgravity conditions on a three-dimensional (3-D) clinostat. Simulated microgravity conditions on a 3-D clinostat did not affect the number of rosette leaves but promoted the growth and development (fresh weight of plant and the elongation of flower stalk) of tran...
متن کاملIn Vitro Growth of Mouse Preantral Follicles Under Simulated Microgravity
14 day-old mouse ovarian tissue and preantral follicles isolated from same-aged mice were incubated in a simulated microgravity culture system. We quantitatively assessed follicle survival, measured follicle and oocyte diameters, and examined ultrastructure of the oocytes produced from the system. We observed decreased follicle survival, downregulation of expressions of proliferating cell nucle...
