Acetyl-11-Keto-β-Boswellic Acid Restrains Inflammation and Extracellular Matrix Degradation of Osteoarthritis via Suppression of NF-κB Pathway

Background: Mechanical stress along with inflammation play causative roles in the development of osteoarthritis (OA), which decreases quality life and causes economic loss. Inflammation extracellular matrix (ECM) degradation have been identified as key factors OA. As main active component frankincense, acetyl-11-keto-β-boswellic acid (AKBA) has shown to positive effects on inflammation. However, AKBA cartilage ECM are currently elusive. Methods: Micro-CT, Histologic analysis, Immunohistochemical (IHC) analysis were conducted vivo Hulth-Telhag OA rat or without treatment. The articular chondrocytes evaluated by CCK-8, Western Blot, quantitative real-time polymerase chain reaction (qRT-PCR), immunofluorescence (IF) assay. We demonstrated role pathogenesis determined protective effect both model lipopolysaccharide (LPS)-induced chondrocytes. Findings: found increased inflammatory expression decreased LPS-induced chondrocytes. Meanwhile, its inhibitory well ECM-related markers also observed model. Furthermore, activation NF-κB attenuated nuclear p65 protein levels upon LPS stimulation. In addition, was subsequently reversed NF- κB signal inflammation-related chondrocytes. Interpretation: Suppression pathway restrains via inhibition degradation. is a promising therapeutic agent for treatment OA.
