چکیده

Coronaviruses such as SARS-CoV-2, which is responsible for COVID-19, depend on virus spike protein binding to host cell receptors cause infection. The SARS-CoV-2 binds primarily ACE2 target cells and then processed by membrane proteases, including TMPRSS2, leading viral internalisation or fusion with the plasma membrane. It has been suggested, however, that other than may be involved in binding. We have investigated interactions of recombinant versions human epithelial lines express low/very low levels TMPRSS2 a proxy assay interaction cells. A tagged form containing S1 S2 regions bound temperature-dependent manner all lines, whereas region alone receptor-binding domain (RBD) interacted only weakly. Spike associated independently while RBD required presence high interaction. As previously shown bind heparin, soluble glycosaminoglycan, we tested effects various heparins ACE2-independent Unfractionated heparin inhibited an IC50 value <0.05 U/mL, two low-molecular-weight were less effective. mutant protein, lacking arginine-rich putative furin cleavage site, weakly had lower affinity unfractionated wild-type protein. This suggests site might also heparin-binding potentially important glycosaminoglycans heparan sulphate dermatan sulphate, but not chondroitin indicating it one both these surface

Evolutionary Analysis of Mammalian ACE2 and the Key Residues Involved in Binding to the Spike Protein Revealed Potential SARS-CoV-2 Hosts

Introduction: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spilled over to humans via wild mammals, entering the host cell using angiotensin-converting enzyme 2 (ACE2) as receptor through Spike (S) protein binding. While SARS-CoV-2 became fully adapted to humans and globally spread, some mammal species were infected back. The present study evaluated the potential risk of mammals...

Inhibition of SARS Pseudovirus Cell Entry by Lactoferrin Binding to Heparan Sulfate Proteoglycans

It has been reported that lactoferrin (LF) participates in the host immune response against Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV) invasion by enhancing NK cell activity and stimulating neutrophil aggregation and adhesion. We further investigated the role of LF in the entry of SARS pseudovirus into HEK293E/ACE2-Myc cells. Our results reveal that LF inhibits SARS pseudovirus in...

Antimyogenic Effect of SARS-CoV Spike Protein in C2C12 Myoblasts

C2C12 myoblasts serve as well-established model system to study myogenesis, as they fuse to form multinucleated myotubes. Severe acute respiratory syndrome coronavirus (SARS–CoV) spike (S) protein plays a crucial role in viral entry. Exogenous expression of S protein in C2C12 myoblasts inhibits the formation of myotubes. Global changes in gene expression were studied in C2C12 cells expressing S...

Spike protein of SARS-CoV stimulates cyclooxygenase-2 expression via both calcium-dependent and calcium-independent protein kinase C pathways.

We have previously shown that the nucleocapsid protein of SARS-associated coronavirus (SARS-CoV) activated cyclooxygenase-2 (COX-2) expression. In this study, we identified another viral protein, the spike of SARS-CoV, which played an important role in virus-stimulated COX-2 expression after screening all genes from the SARS-CoV genome. We found that an upstream calcium-dependent PKC isozyme PK...

Angiotensin-Converting Enzymes, the Key Components in the Pathogenesis of COVID-19 Infection and their Role in the Interaction of SARS-Cov-2 with Human Host Cells

Introduction: Over the past 20 years, seven coronaviruses have caused more or less severe respiratory diseases in humans. Among these, the most important ones are SARS-CoV and the coronavirus, a similar virus that has created a pandemic called Covid-19 since 2019, belonging to the b-category of beta-coronaviruses called Sarbecovirus. This virus is due to a kind of spike-like structure to the A...
