A Water?Soluble Peptoid Chelator that Can Remove Cu 2+ from Amyloid?? Peptides and Stop the Formation of Reactive Oxygen Species Associated with Alzheimer's Disease

چکیده

Cu bound to amyloid-? (A?) peptides can act as a catalyst for the formation of reactive oxygen species (ROS), leading neuropathologic degradation associated with Alzheimer's disease (AD). An excellent therapeutic approach is use chelator that selectively remove from Cu–A?. This should compete Zn2+ ions (Zn) are present in synaptic cleft while forming nontoxic complex. Herein we describe P3, water-soluble peptidomimetic removes Cu2+ Cu–A? presence Zn and prevent ROS even reductive environment. We demonstrate, based on extensive spectroscopic analysis, although P3 extracts Cu,Zn–A? faster than it Cu, formed complexes kinetic products further dissociate, CuP3 an exclusive stable thermodynamic product. Our unique findings, combined bioavailability peptoids, make drug candidate context AD.

Reactive Oxygen Species, Redox Signaling and Neuroinflammation in Alzheimers Disease: The NF-kappa B Connection

Oxidative stress and inflammatory response are important elements of Alzheimer's disease (AD) pathogenesis, but the role of redox signaling cascade and its cross-talk with inflammatory mediators have not been elucidated in details in this disorder. The review summarizes the facts about redox-signaling cascade in the cells operating through an array of kinases, phosphatases and transcription fac...

Reactive oxygen species: from health to disease.

Upon reaction with electrons, oxygen is transformed into reactive oxygen species (ROS). It has long been known that ROS can destroy bacteria and destroy human cells, but research in recent decades has highlighted new roles for ROS in health and disease. Indeed, while prolonged exposure to high ROS concentrations may lead to non-specific damage to proteins, lipids, and nucleic acids, low to inte...

O 22: Reactive Oxygen Species and Epilepsy

Seizure activity has been proposed to result in the generation of reactive oxygen species (ROS), which then contribute to seizure-induced neuronal damage and eventually cell death. Although the mechanisms of seizure-induced ROS generation are unclear, mitochondria and cellular calcium overload have been proposed to have a crucial role. We aim to determine the sources of seizure-induced ROS and ...

the immunopathogenic role of reactive oxygen species in alzheimer disease.

reactive oxygen species (ros) are produced in many normal and abnormal processes in humans, including atheroma, asthma, joint diseases, cancer, and aging. basal levels of ros production  in  cells  could  be  related  to  several  physiological functions  including  cell proliferation, apoptosis and homeostasis. however, excessive ros production above basal levels would impair and oxidize dna, ...

Metal-dependent generation of reactive oxygen species from amyloid proteins implicated in neurodegenerative disease.

Using a method based on ESR spectroscopy and spin-trapping, we have shown that Abeta (amyloid beta-peptide) (implicated in Alzheimer's disease), alpha-synuclein (implicated in Parkinson's disease), ABri (British dementia peptide) (responsible for familial British dementia), certain toxic fragments of the prion protein (implicated in the transmissible spongiform encephalopathies) and the amylin ...
