A Validated Reverse-Phase High Performance Liquid Chromatography (RP-HPLC) Method for the Quantification of Gamma- Tocotrienol in Tocotrienol Rich Fractions of Crude Palm Oil

Background: Palm oil and its constituents have wide applications in food, cosmetics pharmaceutical industries. The tocotrienol-rich fractions of crude palm drawn greater research interest recent years due to their potent health benefits. Therefore, reliable validated analytical methods are essential for the quantification tocotrienols. Objective: This study aimed develop a simple economical RP-HPLC method gamma-tocotrienol oil. Methods: An Agilent HPLC system supplied with Diode Array detector an auto-injector was used development, wavelength set 295 nm. A reversedphase C18 column maintained at 30 °C using mobile phase composition methanol: water (95:05) flow rate 1 mL/min analysis. developed according ICH guidelines. Results Discussion: symmetrical peak observed 8.7 minutes minimal tailing (between 0.76 0.78), acceptable resolution above 2.0. Excellent linearity evident R2 values 0.9996 0.9991 intra-day interday, respectively. demonstrated high precision (%RSD ≤ 5.8%) accuracy (%RE<9.6%). LOD LOQ were determined as 1.4 μg/mL 4.2 μg/mL, suitability studies indicate that chromatographic parameters well within limit. Conclusion: In conclusion, is rapid, precise, stable gamma-tocotrienol.
