Alternate gram staining technique using a fluorescent lectin.

Fluorescence-labeled wheat germ agglutinin binds specifically to N-acetylglucosamine in the outer peptidoglycan layer of gram-positive bacteria. The peptidoglycan layer of gram-negative bacteria is covered by a membrane and is not labeled by the lectin. By exploiting this phenomenon, an alternative Gram staining technique has been developed.

Automatic Gram staining by a linear conveyor system.

The examination of a well-stained film of either clinical material or organisms grown in laboratory media constitutes an important part of most investigations in hospital microbiology laboratories. The staining of large numbers of microscope slides, usually by Gram's method, is however timeconsuming, and a number of reports have been made on methods mechanising this procedure. Cremer (1968) and...

A new technique for Gram staining paraffin-embedded tissue.

Five techniques for Gram staining bacteria in paraffin sections were compared on serial sections of pulmonary tissues from eight bacteriological necropsies. Brown and Hopp's method was the most satisfactory for distinguishing Gram-positive and Gram-negative bacteria. However, this method cannot be recommended as the preparations were frequently overstained, and the Gram-negative bacteria were s...

Rapid lipid-staining procedure for paper electrophoretograms.

Paper electrophoresisiswidely used in clinicalmedicine as a screening test for hyperlipidemia. It is a simple, reproducible procedure, but requires 36 h to complete. Aliquota of sera are electrophoresed 16-18 h, paper strips are dried for 30 mm, followedby overnightstaining(1, 2). Recently it was reported that stainingtime could be decreased to 2 h by using Oil Red 0 stain in acetone-water solv...

Bronchoalveolar lavage: a standardized procedure or a technical dilemma?

Bronchoalveolar lavage (BAL) is widely used for analysis of cells and soluble components of the lower respiratory tract. In its infancy, this simple and safe method has been applied as a pure research tool. Meanwhile, "the child has grown up", and its clinical usefulness for routine diagnostic purposes has been recognized worldwide in the various interstitial lung diseases and particularly in o...
