A scalable device-less biomaterial approach for subcutaneous islet transplantation

چکیده

The subcutaneous space has been shown to be a suitable site for islet transplantation, however an abundance of islets is required achieve normoglycemia, often requiring multiple donors. loss due the hypoxic conditions experience during revascularization, resulting in apoptosis. Therefore, reduce therapeutic dosage pre-vascularization pursued. In this study, we highlight biomaterial-based approach using methacrylic acid copolymer coating generate robust pre-vascularized cavity transplantation. We also devised simple, but not-trivial, procedure filling with suspension collagen. show that can support marginal mass rapidly return streptozotocin-induced diabetic SCID/bg mice normoglycemia. Furthermore, immunocompetent Sprague Daley rats remained normoglycemia up 70 days until they experienced graft destabilization as outgrew their implants. This work highlights acid-based biomaterials strategy scalable and doesn't require exogenous cells or growth factors.

Biological and biomaterial approaches for improved islet transplantation.

Islet transplantation may be used to treat type I diabetes. Despite tremendous progress in islet isolation, culture, and preservation, the clinical use of this modality of treatment is limited due to post-transplantation challenges to the islets such as the failure to revascularize and immune destruction of the islet graft. In addition, the need for lifelong strong immunosuppressing agents rest...

Successful subcutaneous pancreatic islet transplantation using an angiogenic growth factor-releasing device.

INTRODUCTION Although the subcutaneous tissue is considered as an attractive site for pancreatic islet transplantation, the success rate has been extremely low. AIMS To use basic fibroblast growth factor (bFGF) to induce neovascularization and sufficient blood flow around the space formed for grafted islets in the subcutaneous tissue to improve the islet survival. METHODOLOGY In the experim...

Percutaneous approach for human islet transplantation.

Two male patients. aged 29 and 36 years. received combined cadaveric kidney-islet grafts for end-stage renal disease secondary to type I diabetes mellitus. The kidneys and pancreas were obtained from multiorgan donor procurement.2 The organs were preserved in UW solution and packed on ice. The pancreas ofthe kidney donor was the source of the primary islet allograft in the 29-year-old patient. ...

Laparoscopic approach for islet cell transplantation.

SubtotaJ pancreatectomy with preservation of the duodenum was performed in two mongrel dogs weighing 12 and 13.5 kg. The islets were separated by collagenase digestion (Boehringer-Mannheim P, Mannheim. Germany 1.5 mg/mL) using the automated method and purified on Eurocollins-Ficoll discontinuous gradients.2 The final preparation comprised approximately 291,000 and 146,000 islets with an average...

Diabetes Is Reversed in a Murine Model by Marginal Mass Syngeneic Islet Transplantation Using a Subcutaneous Cell Pouch Device

BACKGROUND Islet transplantation is a successful β-cell replacement therapy for selected patients with type 1 diabetes mellitus. Although high rates of early insulin independence are achieved routinely, long-term function wanes over time. Intraportal transplantation is associated with procedural risks, requires multiple donors, and does not afford routine biopsy. Stem cell technologies may requ...
