A new polysaccharide isolated from Dendrobium offcinale, stimulates aquaporin-3 expression in human keratinocytes

Polysaccharides are the major components of water extract from Dendrobium officinale, which have been thoroughly studied and found to favorable moisturizing properties on mammalian skin. However, structures polysaccharides skin remains unknown. The aim this study was characterize D. officinale polysaccharides, anti-oxidant activity role promoting effects aquaporin-3 (AQP3). Two crude polysaccharide fractions were extracted using biological fermentation, respectively. further purified by graded alcohol precipitation, finally DOP-S DOP-F obtained Sephacryl S-300 column chromatography. average molecular weights 3.604 × 105 1.027 Da, Monosaccharide units elucidated gas chromatography-mass spectrometry (GC-MS) Fourier Transform infrared spectroscopy (FTIR). At same time, induced (AQP3) expression evaluated immuno-fluorescence analysis. antioxidant activities 1,1-diphenyl-2-picrylhydrazyl (DPPH) ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging capacities, ferric reducing power (FRAP). results showed that fermentation effectively achieved hydrolysis stimulated AQP3 expression.
