A New Multiplex Real-Time RT-PCR for Simultaneous Detection and Differentiation of Avian Bornaviruses

نویسندگان

چکیده

Avian bornaviruses were first described in 2008 as the causative agents of proventricular dilatation disease (PDD) parrots and their relatives (Psittaciformes). To date, 15 genetically highly diverse avian covering at least five viral species have been discovered different bird orders. Currently, primary diagnostic tool is detection RNA by conventional or real-time RT-PCR (rRT-PCR). One drawbacks this usage either specific assays, allowing one particular virus, assays with a broad spectrum, which, however, do not allow for simultaneous specification detected virus. facilitate bornaviruses, multiplex assay was developed. Whole-genome sequences various aligned. Primers designed to recognize conserved regions within overlapping X/P gene probes selected detect virus species-specific target region. The optimization resulted sensitive Psittaciformes, Passeriformes, aquatic birds. Finally, new rRT-PCR successfully employed field samples from species. This will serve powerful epidemiological studies improve bornavirus detection.

Development of multiplex real-time RT-PCR as a diagnostic tool for avian influenza.

A multiplex real-time reverse transcriptase-polymerase chain reaction (RRT-PCR) assay for the simultaneous detection of the H5 and H7 avian influenza hemagglutinin (HA) subtypes was developed with hydrolysis type probes labeled with the FAM (H5 probe) and ROX (H7 probe) reporter dyes. The sensitivity of the H5-H7 subtyping assay was determined, using in vitro transcribed RNA templates, to have ...

Detection of a novel avian influenza A (H7N9) virus in humans by multiplex one-step real-time RT-PCR assay

BACKGROUND A novel avian influenza A (H7N9) virus emerged in eastern China in February 2013. 413 confirmed human cases, including 157 deaths, have been recorded as of July 31, 2014. METHODS Clinical specimens, including throat swabs, sputum or tracheal aspirates, etc., were obtained from patients exhibiting influenza-like illness (ILIs), especially from those having pneumonia and a history of...

Rapid detection and serotyping of dengue virus by multiplex RT-PCR and real-time SYBR green RT-PCR.

INTRODUCTION Dengue fever and dengue haemorrhagic fever currently rank highly among the newly-emerging infectious diseases, and are considered to be the most important arboviral disease worldwide. The definitive diagnosis is culture analysis, but practical considerations limit its use. Also, the period for viral detection is limited. Within a day or two after fever subsides, rising levels of an...
