چکیده

Breast cancer (BC), is a heterogeneous disease, divided into molecular subtypes based on gene expression and clinical outcomes. MABC poorly diagnosed subtype, characterized by estrogen (ER), progesterone receptor (PR) negativity, HER-2-amplification in most cases; yet it overexpresses androgen (AR) activates its pathway. Its improper diagnosis demands the adoption of complementary tools to refine classification; miRNA, key players regulating cell behavior, AR, tumorigenic processes BC hold promise defining studying behavior MABC. 539 breast microarray data were downloaded from The Cancer Genome Atlas (TCGA-BRCA). Cases with available miRNA retained classified using citbcmst R package (PMID: 30289602). Differential analysis identified deregulated miRNAs DEseq2. A validation set 131 ER-neg samples was constituted tumors apart triple-negative (TN)BC previously described signature (AR, FOXA1 AR-related genes, PMID: 23663520) fresh tissue sections. Using miRCURY LNATM PCR assay, profiling done for panel 6 differentially expressed miRNAs. Finally, silico predicted target genes these assessed RT-qPCR TNBC lines after transfection mimics. TCGA indicated as separate entity signature. MiRNA-seq depicted significantly absolute value log2 fold change> 1 P-adjusted <0.05 between TNBC. We validated 78 (27 HER2-/ 51 HER2+), 53 TN samples, profiling, significant upregulation miR-2115-3p miR-187-3p compared Preliminary data, showed that overexpression downregulates (FGF2, HMGCS1, ADAM12) implicated carcinogenesis differently each models used. has unique regulate can potentially serve biomarkers effectively diagnose, prognose treat

The Clinicopathologic Features of Molecular Apocrine Breast Cancer

BACKGROUND To elucidate the clinicopathologic features and their implications on the immunohistochemistry in cases of molecular apocrine breast cancer (MABC). METHODS Immunohistochemical (IHC) staining for estrogen receptor (ER), human epidermal growth factor receptor 2 (HER-2), cytokeratin (CK) 5/6, epidermal growth factor receptor (EGFR), androgen receptor (AR), gamma-glutamyltrasferase 1 (...

p62 Regulates the Proliferation of Molecular Apocrine Breast Cancer Cells

p62, also called sequestosome 1 (SQSTM1), is a multifunctional signaling molecule that affects cell proliferation. Recently, we found accumulation of p62 in apocrine carcinoma of the breast, however, the biological role of p62 expression in apocrine carcinoma still remains unclear. To investigate whether p62 might contribute to tumor cell proliferation in apocrine carcinomas, we used the MDA-MB...

HER2 status in molecular apocrine breast cancer: associations with clinical, pathological, and molecular features.

Molecular apocrine breast cancer (MABC) is a distinct subtype of breast cancer. The purpose of this study was to investigate the relationship between HER2 status and clinicopathologic characteristics of MABCs from Chinese Han cohort. A cohort of 90 MABC patients were enrolled. Immunohistochemical method was performed to analyze the molecular expression, and the human epidermal growth factor rec...

Basal Breast Cancer: A Complex and Deadly Molecular Subtype

During the last decade, gene expression profiling of breast cancer has revealed the existence of five molecular subtypes and allowed the establishment of a new classification. The basal subtype, which represents 15-25% of cases, is characterized by an expression profile similar to that of myoepithelial normal mammary cells. Basal tumors are frequently assimilated to triple-negative (TN) breast ...
