Abstract Background and Aims The ADR mouse model is a well-established rodent of FSGS that can manifest with albuminuria. Resident macrophages are present in the tubulointerstitium kidney, circulating be recruited to kidney disease states. However, presence glomeruli role podocyte injury models remain unknown. Method Single-cell RNA sequencing comprehensively characterized expression mice glomeruli. Eight-week-old male BALB/c were used induce model. Immunofluorescence was detect changes renal after injected at different time points. Transwell co-culture system effect Lrg1+Vcan+Ly6Chigh on MPCs. HCH6-1,the FPR1 inhibitor restrain ANXA1-FPR1 pathway. Results Totally 506 sequenced significantly increased induced nephrotic (P<0.01). ligand receptor relationship between podocytes changed(P<0.01, means>1). demonstrated glomeruli(P<0.05). temporal spatial still need proved by further experiments. After Lrg1+Vcan+Ly6Chighmacrophages MPCs, nephrin MPCs decreased(P<0.05), which cause injury. In group, signaling pathway activated, Lrg1+Vcan+Ly6Chighmacrophage infiltration, albuminuria reduced ADR+HCH6-1gruop (all P<0.05). Conclusion into activation damage podocyte.
