چکیده
Current therapeutic options for atopic dermatitis (AD) consist of either broad or targeted immunosuppressive agents. However, the natural course AD can hardly be modified, as disease invariably returns after cessation treatment. Tissue-resident memory T-cells are hypothesized to relevant players in mediating disease-specific ‘immune memory’, but their exact immunopathological phenotype is so far unknown. By using a multi-omics approach involving single-cell RNA sequencing combined with multiplex proteomics skin samples, we studied patients undergoing short (16 weeks) and long-term (one year) treatment IL-4Rα blocker dupilumab. blockade resulted clearance disease, decrease immune cell counts, normalization transcriptomic dysregulation keratinocytes. Interestingly, found distinct populations dendritic cells (DC) that were largely absent healthy control persist up one year These included LAMP3+ CCL22+ mature DC, CRTH2+ CD161+ Th2A cells, CRTAM+ cytotoxic T-cells, expressing peak levels CCL17 IL13 (T-cells). showed specific receptor constellation IL17RB, IL1RL1 (ST2) CRLF2, possibly rendering them key responders AD-typical epidermal alarmins IL25, IL33 TSLP. We thus identified persisting DC maintained an inflammatory treatment, equipped all receptors facilitate keratinocyte-DC-Th2-mediated response. emerge central skin-intrinsic leads recurrences, might therefore promising targets achieve more sustained

Experimental atopic dermatitis depends on IL-33R signaling via MyD88 in dendritic cells
Atopic dermatitis (AD) is a chronic Th2 type inflammatory skin disorder. Here we report that MyD88 signaling is crucial in the pathogenesis of experimental AD induced by vitamin D3 analog MC903. The clinical signs and inflammation caused by MC903 are drastically reduced in MyD88-/- mice with diminished eosinophil, neutrophil infiltration and Th2 cytokine expression. The biological effect of int...

Background: Atopic dermatitis is a chronic, recurrent inflammation of the skin, accompanied by severe pruritus. Immune system dysregulation and skin barrier defects are associated with the abnormalities in atopic dermatitis. Myriad pieces of evidence have pointed to the major roles of LL-37 and interleukin-31 (IL31)in atopic dermatitis. The studies on atopic dermatitis are still limited i...

Atopic dermatitis (AD) is an inflammatory disorder of the skin characterized by an impaired immune response. Several effector T cell subsets, such as pro-inflammatory cells like Th9, Th17 and Th22 cells, expressing high levels of IL-9, IL-17 and IL-22, together with the anti-inflammatory, immuno-modulating Treg cells constitutively producing IL-10, seem to play a role in this condition. IL-9 an...
متن کاملGeneration of Mature Monocyte-Derived Dendritic Cells in the Presence of Heparin and Monocyte Conditioned Medium: Phenotypic and Functional Comparison
Background: Dendritic cells (DC) induce tumor or pathogen-specific T cell responses in humans. Several laboratories have developed culture systems, including maturation factors for human DC from peripheral blood monocytes. We comprehensively compared standard maturation stimulus, an autologous monocyte-conditioned medium (MCM), with heparin for their ability to promote uniformly mature DC that ...

IMPORTANCE Atopic dermatitis (AD) is a common illness of childhood. OBJECTIVE To evaluate the natural history of AD and determine the persistence of symptoms over time. DESIGN, SETTING, AND PARTICIPANTS A cross-sectional and cohort study of a nation-wide long-term registry of children with AD enrolled in the Pediatric Eczema Elective Registry (PEER). MAIN OUTCOMES AND MEASURES Self-report...
