560 Isolation of an “early” transit amplifying keratinocyte population in interfollicular human epidermis: a role for CD271 receptor

چکیده
In the interfollicular epidermis stem cells (KSC) generate transit amplifying (TA) that, after symmetric divisions, produce differentiating daughters. We isolated and characterized highly proliferative epidermal basal cell population “early” TA (ETA) cells, based on its capacity to adhere type IV collagen. Proliferation colony forming efficiency in ETA are lower than KSC, but higher “late” (LTA). Stem differentiation markers confirmed that display a unique phenotype. Skin reconstructs derived from present different features (epidermal thickness, Ki67 survivin expression), as compared skin equivalents generated either KSC or LTA cells. The low affinity neurotrophin receptor CD271, regulates transition human through an on/off switch control mechanism, is predominantly expressed siRNA CD271 more less differentiated phenotype, mock-derived reconstructs. Consistently, overexpression generates equivalent mock Finally, expression declines with ageing cellular senescence. addition, overexpressing show p16ink4a levels during replicative senescence also appears delay comparison conclude represent first progenitor exclusive features. identifies modulates thus participating early regenerative of epidermis.

Mitotic Diversity in Homeostatic Human Interfollicular Epidermis
Despite decades of skin research, regulation of proliferation and homeostasis in human epidermis is still insufficiently understood. To address the role of mitoses in tissue regulation, we utilized human long-term skin equivalents and systematically assessed mitoses during early epidermal development and long-term epidermal regeneration. We now demonstrate four different orientations: (1) horiz...
متن کاملRole of melanoma chondroitin sulphate proteoglycan in patterning stem cells in human interfollicular epidermis.
Human interfollicular epidermis is renewed by stem cells that are clustered in the basal layer in a patterned, non-random distribution. Stem cells can be distinguished from other keratinocytes by high expression of beta1 integrins and lack of expression of terminal differentiation markers; they divide infrequently in vivo but form actively growing colonies in culture. In a search for additional...

15 صفحه اولTLR7-expressing cells comprise an interfollicular epidermal stem cell population in murine epidermis
Normal interfollicular epidermis (IFE) homeostasis is maintained throughout the entire life by its own stem cells that self-renew and generate progeny that undergo terminal differentiation. However, the fine markers of the stem cells in interfollicular epidermis are not well defined yet. Here we found that TLR7 identified the existence of progenitors and interfollicular epidermal stem cells in ...
متن کاملIsolation and Cultivation of Adult Human Keratinocyte Stem Cells for Regeneration of Epidermal Sheets
Background: Keratinocyte stem cell is one of the adult stem cells that inhabits the skin and contributes to skin function and renewal. Adult stem cells are best defined by their capacity to self-renew, and to maintain tissue function for a long period of time. These findings indicate the importance of these cells for clinical applications including regenerative medicine, tissue engineering and ...
