357 Cutibacterium acnes CAMP factor 1-related peptides as Toll-Like Receptor-2 modulators

Acne vulgaris is a disorder affecting the pilosebaceous unit resulting in both inflammatory and non-inflammatory clinical lesions. Cutibacterium acnes (C. acnes) of specific lineage, plays key role acne by producing several virulence factors thereby increasing their capacity. The C. mutated CAMP1 factor display strong TLR-2 binding property highly strains. peptide array analysis 116 15-mer peptides sequences with an overlap 11 amino acids, were immobilized onto glass plate showed that seven (A14, A15, B1, B2, B3, C1 C3) specifically recognized located on same side silico 3D-CAMP1 structure. Among these peptides, 2 (A14 B2) able to compete interaction CAMP1-TLR-2 vitro. Moreover, 3 B1 dose-dependently decrease acnes-induced CX-CL8 production keratinocytes as well at transcriptional translational levels ex vivo GM-CSF, TNF-α, IL-1β, IL-10 human skin explants stimulated recombinant (r)CAMP1 protein. RNA sequencing pretreated B2 rCAMP1, identified 10 genes, IL6, TNF, CXCL1, CXCL2, CXCL3, CXCL8, CCL2, CCL4 CSF2, harboring differential expression. All genes validated qRT-PCR. We characterized new CAMP1-derived which could be used potential modulator reduce TLR-2-induced inflammation.
