چکیده

Psoriasis is an inflammatory disorder which leads to the improper stratification of epidermal keratinocytes. Autophagic flux analysis our established cell lines has shown that autophagy functionally active in healthy keratinocytes but impaired psoriatic To mirror psoriasis vitro we custom-made a cytokine cocktail induced IL-1β release keratinocyte cultures. Stimulation by this chronic similarly causes block and accumulation key proteins. Interestingly, stimulus LC3-I accumulation, rather than LC3-II, suggesting inflammation blocks prior final stages autophagosome autolysosome formation. In addition, p62 upregulation indicates disruption cargo degradation via lysosomal compartment. Increased ATG4, ATG7 ATG12 expression these were not cause pathway. Changes phospholipid metabolism been reported skin conditions including Phosphatidylethanolamine (PE) downregulation PE essential for conversion LC3-II. By inducing production inflamed keratinocytes, show restoration pathway functionality. Transcriptomic data identified several autophagic genes with significantly deregulated samples (n=398) compared matched or non-matched (n=587) controls. Immunofluorescence staining human confirmed differences marker deregulation membrane We have impairs downregulating altering compartment integrity. Our supports stimulation as beneficial approach counteract such psoriasis.

Chronic pain leads to concomitant noradrenergic impairment and mood disorders.

BACKGROUND Patients suffering chronic pain are at high risk of suffering long-lasting emotional disturbances characterized by persistent low mood and anxiety. We propose that this might be the result of a functional impairment in noradrenergic circuits associated with locus coeruleus (LC) and prefrontal cortex, where emotional and sensorial pain processes overlap. METHODS We used a chronic co...

Upregulation of ANGPTL6 in mouse keratinocytes enhances susceptibility to psoriasis

Psoriasis is a chronic inflammatory skin disease marked by aberrant tissue repair. Mutant mice modeling psoriasis skin characteristics have provided useful information relevant to molecular mechanisms and could serve to evaluate therapeutic strategies. Here, we found that epidermal ANGPTL6 expression was markedly induced during tissue repair in mice. Analysis of mice overexpressing ANGPTL6 in k...

Evaluation of the therapeutic response to phototherapy at different body sites in chronic plaque-type psoriasis

Background and aim: Psoriasis is a chronic inflammatory dermatosis that affects 0.6% to 4.8% of the population. Phototherapy is commonly used in the treatment of this disease. According to the results of previous studies that showed difference in minimal erythema dose between different body sites, it seems that the responses of psoriatic lesions are also different in various body sites. O...

The role of Staphylococcus superantigens in chronic plaque type psoriasis

Background: T cell activation is discussed as an important factor in the pathogenesis of psoriasis. Recently, a lot of attention has been paid to the role of superantigens in T cell activation in the pathogenesis of psoriasis. In this study, the role of staphylococcal superantigens in the pathogenesis of psoriasis with identification of staphylococcal toxin levels in the skin lesions of p...

Keratinocytes contribute intrinsically to psoriasis upon loss of Tnip1 function.

Psoriasis is a chronic inflammatory skin disease with a clear genetic contribution, characterized by keratinocyte proliferation and immune cell infiltration. Various closely interacting cell types, including innate immune cells, T cells, and keratinocytes, are known to contribute to inflammation. Innate immune cells most likely initiate the inflammatory process by secretion of IL-23. IL-23 medi...
