208 Single Cell RNA Sequencing (Scrna-seq) in Studying the Early Embryonic Muscle Development

Abstract During the early development, skeletal muscle, adipose, and connective tissues have high plasticity are sensitive to in uterine nutritional fluctuations. past decade, maternal nutrition-induced changes myogenesis adipogenesis during fetal stage been extensively studied their alterations closely associated with production efficiency meat quality of farm animals. Organogenesis initiates embryonic stage, which sets blueprint for later growth development. Limited but growing evidence support that also profound effects on animal efficiency. Using single-cell transcriptomic analysis (scRNA-seq), we investigated obesity (MO) day 9.5 (E9.5) mouse embryos. First, identified myogenic subpopulations within dermomyotome (DM) a developmental trajectory was reconstructed. Our results showed MO induced systematic hypoxia, correlated enhanced BMP signaling impaired muscle differentiation DM. The Notch-signaling effectors, HES1 HEY1, act downstream signaling, were activated by hypoxia-inducible factor 1 (HIF1A) suppressed through transcriptionally repressing an important regulator MEF2C. In conclusion, induces hypoxia impairs up-regulating DM, may account disruptions development function progeny. Understanding programming as well adipose fibrotic tissues, especially at will facilitate stage-specific management strategies optimize quality.
