چکیده

The antimicrobial peptide derived from insulin-like growth factor-binding protein 5 (AMP-IBP5) displays antimicrobialactivities and immunomodulatory properties; however, its role in the regulation of skin barrier context atopic dermatitis (AD) remains unclear. Normal human keratinocytes were stimulated with AMP-IBP5 Western blot was used to analyze expression tight junction (TJ)-related proteins. Immunofluorescence staining examine intercellular distribution TJ-related Activation signaling pathways aPKC Rac1 evaluated by TJ function investigated measurement transepithelial electrical resistance (TER). BALB/c mice treated 2,4-dinitrochlorobenzene as AD mouse model. permeability assay immunofluorescence investigate vivo mice. We found that increased various proteins, including claudin-1, occludin zonula occludens 1 enhanced their along cell-cell borders. also improved shown increasing TER keratinocyte. In addition, phosphorylation Rac1, which are required for AMP-IBP5-mediated improvement barrier, evidenced inhibitory effects respective inhibitors on TER. mice, symptoms, recovered proteins function. Additionally, suppressed production inflammatory cytokines such IL-4, IL-13, IL-31, IL-33 TSLP. finding relieves symptoms enhances indicates a possible application treatment AD.

Skin barrier in atopic dermatitis.

The skin represents the largest organ of the body and provides a vital interface between the body and the environment. Hereditary and acquired alterations of structural proteins and lipids of the stratum corneum and epidermal tight junctions leading to a diminished skin barrier function are major causative factors for a number of skin diseases, in particular atopic dermatitis (AD). This review ...

Skin barrier in atopic dermatitis.

Research about the skin barrier and its properties has increased significantly since the 60s, with studies that indicated its resistance when isolated, as well as its particularities in relation to skin permeability. At the same time, description of Odland bodies helped to understand how stratum corneum stability is maintained. The âbrick and mortarâ model is the most accepted so far. In this a...

TRPV1 antagonist can suppress the atopic dermatitis-like symptoms by accelerating skin barrier recovery.

BACKGROUND Transient receptor potential vanilloid type 1 (TRPV1) is a cation channel activated by diverse obnoxious stimuli like capsaicin, low pH or heat. Recently, it was revealed that TRPV1 might be deeply associated with skin permeability barrier function, suggesting that modulation of TRPV1 might be beneficial for the skin disorders with barrier damages. OBJECTIVE We aimed to investigate...

Insulin-like growth factor I and insulin-like growth factor binding protein 5 in Crohn's disease.

Insulin-like growth factor (IGF)-I and its binding protein IGF binding protein 5 (IGFBP-5) were highly expressed in inflamed and fibrotic intestine in experimental Crohn's disease. IGF-I induced proliferation and increased collagen synthesis by smooth muscle cells and fibroblasts/myofibroblasts in vitro. Here we studied IGF-I and IGFBP-5 in Crohn's disease tissue. Tissue was collected from pati...

Purpose: This study investigated the effect of endurance activity on brain-derived neurotrophic   factor(BDNF), insulin-like growth factor 1(IGF-1) and its binding protein 3(IGFBP-3) in elderly   healthy individuals.   Materials and Methods: Eleven healthy old males (mean age of 68 ± 2.31 years old, height   of 177 ± 3.1 cm and weight of 79 ± 1.5 kg) were studied. Seventy two hours after maxi...
