Psoriasis is an immune-mediated systemic skin disorder with a strong genetic predisposition. The susceptibility gene nitric oxide synthase 2 (NOS2) specifically upregulated in psoriatic involved compared to different types of eczema. However, the functional relevance NOS2 psoriasis unknown. Here, we detected mRNA expression epidermis but were unable verify its overexpression using immunohistochemistry, suggesting marginally epidermal NO. We found that psoriasis-associated miRNAs miR-31 and miR-146a prevented full translation keratinocytes demonstrated this effect was IL-17 dependent. Furthermore, showed induced through RAC1 NOS2-generated NO mediated downstream effects. Interestingly, donor DETA-NONOate range 0.1-1μM, observed significantly increased keratinocyte proliferation colony formation, whereas levels 500-1000 μM led reduced proliferation. therefore hypothesized inflammation may be alleviated by either eliminating or increasing cellular levels. Using imiquimod mouse model, phenotype IMQ- treated KO mice as well IMQ NO-releasing berdazimer cream SB414. In conclusion, induces fine tunes towards window proinflammatory hyperproliferative effects support therapy new treatment modality for psoriasis.
