1542 Allergen sensitization status stratifies IL-31 production by memory T cells in atopic dermatitis

The role of allergen in T-cell mediated IL-31 production and its relationship with clinical features atopic dermatitis (AD) is unknown. by T cells has been mainly analysed qRT-PCR intracellular flow cytometry polyclonal activated peripheral blood mononuclear a low number patients without association the clinic. We studied memory response to house dust mite (HDM) when cocultured autologous lesional epidermal from 45 adult non-treated moderate-to-severe AD 9 control subjects using xMAP technology. Plasma mRNA expression skin lesions were also assessed. defined two groups based on HDM-induced CLA+ cells. In producing group, correlated patient’s pruritus (r=0.57, p=0.045), plasma CCL27 (r=0.60, p=0.032) periostin (r=0.56, p=0.050). Patients group younger, showed more inflammatory profile, higher HDM-specific (sp) total IgE levels than non-producing group. More, sp >100 kUA/L increased vitroresponse HDM, which SCORAD (r=0.61, p=0.040), periostin, CCL18, CCL22 sIL-2R compared those <100 kUA/L. Our findings point between sensitization status that can help understand patients’ heterogeneity IL-31-directed therapies.
