137 Acute stress aggravates itch in experimental AD and drives changes in trigeminal ganglion neurons related to neuroinflammation

Itch is often exacerbated by psychological stress in patients with atopic dermatitis (AD). However, how influence AD-associated itch and the underline mechanisms remain largely unknown. To investigate effect of on behaviors AD-like mice model, we employed a MC903 mouse model AD exposed both vehicle-treated MC903-treated C57BL/6 to 1-hour restraint daily for 7 days. On day 2, observed approximately 1.5-fold increase scratching bouts stressed MC903-AD compared non-stressed that received same treatment. While, repeated more than 4 days combined treatment showed reduced general activity, suggesting an increased anxiety. In order characterize transcriptional changes occur trigeminal ganglia (TG) primary sensory neurons response acute stress, performed RNA-Seq analysis or without 2 There were 2625 differentially expressed genes between TGs from vehicle controls 896 (fold change > 2; false discovery rate< 0.05). Pathways upregulated control included those related synaptic organization transmission, trans-synaptic signaling, axon development, ion binding. Acute caused upregulation neuropeptides (Nts, Npy, Nppb), neutrophil activation (Mpo, Elane), mast cell (Mcpt8, Cpa3), type 1-and 2-inflammation (Il12a, Tslp, Il5) TGs. display elevated (Ngp, Camp, Lcn2) voltage-gated channel (Kcne3) mice. From these results can uncover potential targets controlling stress-aggravated AD.
