1353 Contribution of YBX1 phosphorylation in human epidermal stem cell aging

چکیده

The RNA binding protein YBX1 is a key regulator of epidermal progenitor cell senescence that prevents the mRNA translation critical senescence-associated secretory phenotypes (SASP) transcripts such as CXCL1 and IL8. Decreased levels during aging enables production SASP-related proteins subsequent induction senescence. Posttranslational modifications YBX1, phosphorylation at Ser 102, can modify intracellular localization diminish its capacity. In this study, we investigated role in human skin aging. While MCSP-positive stem/progenitor cells with YBX-1 expression be found abundantly young sun-protected skin, their numbers photoaged decrease. addition, level phosphorylated was increased. Since by PI3 kinase, kinase inhibitors PI-103 GDC-0941 restrict prevent keratinocyte ex vivo cultured increased cells. promising cosmetic ingredient suppresses phosphorylation, decreases IL-8 production, maintains stem conclusion, contributes to aging, inhibition novel anti-aging approach maintaining healthy

The Role of p16INK4a Pathway in Human Epidermal Stem Cell Self-Renewal, Aging and Cancer

The epidermis is a self-renewing tissue. The balance between proliferation and differentiation processes is tightly regulated to ensure the maintenance of the stem cell (SC) population in the epidermis during life. Aging and cancer may be considered related endpoints of accumulating damages within epidermal self-renewing compartment. p16INK4a is a potent inhibitor of the G1/S-phase transition o...

Isolation and Cultivation of Adult Human Keratinocyte Stem Cells for Regeneration of Epidermal Sheets

Background: Keratinocyte stem cell is one of the adult stem cells that inhabits the skin and contributes to skin function and renewal. Adult stem cells are best defined by their capacity to self-renew, and to maintain tissue function for a long period of time. These findings indicate the importance of these cells for clinical applications including regenerative medicine, tissue engineering and ...

Patterning as a signature of human epidermal stem cell regulation.

Understanding how stem cells are regulated in adult tissues is a major challenge in cell biology. In the basal layer of human epidermis, clusters of almost quiescent stem cells are interspersed with proliferating and differentiating cells. Previous studies have shown that the proliferating cells follow a pattern of balanced stochastic cell fate. This behaviour enables them to maintain homeostas...

Comparative genomics of human stem cell factor (SCF)

Stem cell factor (SCF) is a critical protein with key roles in the cell such as hematopoiesis, gametogenesis and melanogenesis. In the present study a comparative analysis on nucleotide sequences of SCF was performed in Humanoids using bioinformatics tools including NCBI-BLAST, MEGA6, and JBrowse. Our analysis of nucleotide sequences to find closely evolved organisms with high similarity by NCB...

Autophagy in stem cell aging

Aging is responsible for changes in mammalian tissues that result in an imbalance to tissue homeostasis and a decline in the regeneration capacity of organs due to stem cell exhaustion. Autophagy is a constitutive pathway necessary to degrade damaged organelles and protein aggregates. Autophagy is one of the hallmarks of aging, which involves a decline in the number and functionality of stem ce...
