چکیده

Microphthalmia-associated transcription factor(MITF) is a key player in phenotype switching melanoma. We showed previously that MITF regulates phenotypic melanoma heterogeneity through changing extracellular matrix(ECM) composition and tumor microarchitecture. now hypothesize controls immune cell infiltration changed ECM structure our 3D spheroid model. Our results demonstrate can impact infiltration, yet this dependent on line additional factors, including chemokines cytokines, disease stage (i.e. primary vs. metastatic). To investigate influence from chemokine secretion, we investigated levels the ECM, found MITF-high lines possessed significantly higher IL-8 than MITF-low lines. Notably, peripheral blood mononuclear cells (PBMCs) preferably killed proliferating G1 phase of cycle, whereas G1-arrested escaped surveillance. However, phenomenon appears to be will need tested multiple different stages. Nevertheless, Natural Killer (NK) are likely responsible for elimination, as short duration experiment does not allow T activation. Interestingly, similar elimination was observed when treated with tumor-specific cells. Taken together, firstly reveal correlation between which potentially support predictor clinical outcomes, linked checkpoint inhibitor therapy outcome overall survival patients. Secondly, report first time PBMCs may prefer proliferative phase. This major challenge current treatments, dormant often relapse. Moreover, finding significant beyond melanoma, occurs range solid tumors, prostate, breast, ovarian lung cancer.

CCL21-induced immune cell infiltration.

Cellular immune responses can be initiated via peptide presentation by specialized antigen presenting cells, dendritic cells (DCs), which stimulate naïve T cells. The trafficking of DCs and T cells is regulated by chemokines such as CCL21. CCL21 is normally expressed in the lymphoid organs and coordinates the interactions between DCs and T cells, thereby contributing to the initiation of T cell...

The Changes of Angiogenesis and Immune Cell Infiltration in the Intra- and Peri-Tumoral Melanoma Microenvironment

Malignant melanoma (MM) urgently needs identification of new markers with better predictive value than currently-used clinical and histological parameters. Cancer cells stimulate the formation of a specialized tumor microenvironment, which reciprocally affects uncontrolled proliferation and migration. However, this microenvironment is heterogeneous with different sub-compartments defined by the...

Human melanoma metastases demonstrate nonstochastic site-specific antigen heterogeneity that correlates with T-cell infiltration.

PURPOSE Metastasis heterogeneity presents a significant obstacle to the development of targeted cancer therapeutics. In this study, we sought to establish from a large series of human melanoma metastases whether there exists a determined pattern in tumor cellular heterogeneity that may guide the development of future targeted immunotherapies. EXPERIMENTAL DESIGN From a cohort of 1,514 patient...

Phenotypic heterogeneity of melanoma. Relation to the differentiation program of melanoma cells

Phenotypic heterogeneity is a characteristic feature of tumor lesions in patients with melanoma. Variability can be observed in cell morphology, pigmentation, and antigen expression. To test whether phenotypic heterogeneity could be the result of events regulated during cell differentiation, we evaluated the expression of a panel of differentiation traits on melanoma cells. Metastatic melanoma ...
