077 Identifying genetic factors that enable basal cell carcinoma to transition from microscopic to macroscopic disease

چکیده
Basal cell carcinoma (BCC) is driven by constitutive activation of the Hedgehog (Hh) signaling pathway, most commonly through loss-of-function mutations in PTCH1. Using animal models, we demonstrate here that microscopic BCC-like tumors initiated loss Ptch1 or gain Smo fail to progress and eventually enter into a dormant state. Because NOTCH1/2 TRP53 are detected human BCC, sought determine if these genetic factors modulate tumor progression. Indeed, found concomitant either Notch1 Trp53 enables rare macroscopic form. whole exome sequencing IHC, further “successful” acquired ability hyperactivate downstream Hh amplifying Gli1/2 upregulating Mycn. Finally, transgenic overexpression MYCN promotes progression disease. Overall, provide functional evidence BCCs arise step-wise process activate upstream signaling, followed acquisition secondary signaling. Our observations may explain why some persist during pharmacological inhibition SMO highlight as potential target for therapy.

Modeling tumor cell migration: From microscopic to macroscopic models.
It has been shown experimentally that contact interactions may influence the migration of cancer cells. Previous works have modelized this thanks to stochastic, discrete models (cellular automata) at the cell level. However, for the study of the growth of real-size tumors with several million cells, it is best to use a macroscopic model having the form of a partial differential equation (PDE) f...

متن کاملFrom Microscopic to Macroscopic Traffic Models
The paper presents a systematic derivation of macroscopic equations for freeway traffic flow from an Enskog-like kinetic approach. The resulting fluid-dynamic traffic equations for the spatial density, average velocity, and velocity variance of vehicles are compared to equations, which can be obtained from a microscopic force model of individual vehicle motion. Simulation results of the models ...
متن کاملValue of CD10 Expression in Differentiating Cutaneous Basal from Squamous Cell Carcinomas and Basal Cell Carcinoma from Trichoepithelioma
Background: In addition to the well-defined histological criteria for squamous cell carcinoma (SCC) and basal cell carcinoma (BCC), immunohistochemical techniques can be used in difficult cases for their differentiation. As differential diagnosis between trichoepithelioma (TE) and BCC is sometimes difficult for the clinician and the pathologist, CD10 may be a useful marker for definite diagnosi...
متن کاملSecondary Involvement of the Mandible due to Basal Cell Carcinoma: A Case Report
Basal cell carcinoma (BCC) is the most common cutaneous malignancy among Caucasians. Rare examples of aggressive and neglected BCC have been reported. Here we report a unique case of a neglected BCC with significant jaw involvement.A 50-year-old female, referred by an otorhinologist, presented with a large ulcer on her chin, which was extended to her mandibular vestibule. The ulcer was 9×5.5 cm...
