064 CD301b+ dermal dendritic cells drive activation of cytotoxic T lymphocytes in a murine contact dermatitis

چکیده
Dermal dendritic cells (DCs) have been found to activate cytotoxic T lymphocytes (CTLs) within inducible skin-associated lymphoid tissue (iSALT) in a murine contact hypersensitivity (CHS) model. DCs are further classified into conventional DC1 (cDC1), DC2 (cDC2), and monocyte-derived DC (MoDC). Furthermore, CD301b+ cDC2, subpopulation of resides specifically non-lymphoid barrier tissues including the skin. However, which subset activates CTL during CHS elicitation phase is unclear. To clarify this, we used XCR1-diphtheria toxin receptor (DTR) CD301b-DTR mice deplete cDC1 respectively. remove MoDC, an anti-CCR2 depleting antibody. Mice were sensitized with 0.5% DNFB (2,4-dinitrofluorobenzene) on abdominal skin (day 0). Following administration diphtheria or antibody, challenged 0.3% ears 5). We that ear swelling interferon-gamma (IFN-γ) production from significantly attenuated by depletion cDC2 MoDC day 6, while did not affect response. Immunohistochemical analysis revealed iSALT formation was only suppressed cDC2. Besides, colocalized clusters. In addition, expression level CD80, costimulatory molecule contributes cell activation, upregulated compared other subsets. Finally, co-culture DNFB-sensitized enhanced IFN-γ 72 hours after stimulation sulfonic acid. Taken together, newly report but cDC1, known present antigens lymph nodes, forming phase. The interaction between may provide novel therapeutic target for CTL-mediated diseases.

Direct activation of CD8+ cytotoxic T lymphocytes by dendritic cells
Recent experiments (11-13) have shown that antigen-specific, CD8+, CD4- T lymphocytes can be induced to proliferate and become killer cells in the absence of a second population of "helper" CD8-, CD4+ cells. We have studied early events in the activation of CD4+ and CD8+ T cell subsets in the primary mixed leukocyte reaction. Dendritic cells are a major if not essential accessory cell for the a...
متن کاملNociceptive Sensory Fibers Drive Interleukin-23 Production from CD301b+ Dermal Dendritic Cells and Drive Protective Cutaneous Immunity.
Innate resistance to Candida albicans in mucosal tissues requires the production of interleukin-17A (IL-17A) by tissue-resident cells early during infection, but the mechanism of cytokine production has not been precisely defined. In the skin, we found that dermal γδ T cells were the dominant source of IL-17A during C. albicans infection and were required for pathogen resistance. Induction of I...
متن کاملActivation of antitumor cytotoxic T lymphocytes by fusions of human dendritic cells and breast carcinoma cells.
We have reported that fusions of murine dendritic cells (DCs) and murine carcinoma cells reverse unresponsiveness to tumor-associated antigens and induce the rejection of established metastases. In the present study, fusions were generated with primary human breast carcinoma cells and autologous DCs. Fusion cells coexpressed tumor-associated antigens and DC-derived costimulatory molecules. The ...

The priming of an immune response against a major histocompatibility complex class I-restricted antigen expressed by nonhematopoietic cells involves the transfer of that antigen to a host bone marrow-derived antigen presenting cell (APC) for presentation to CD8+ T lymphocytes. Dendritic cells (DC), as bone marrow-derived APC, are first candidates for presentation of tumor-associated antigens (T...
