چکیده
Lichen planus (LP) and discoid lupus erythematosus (DLE) are chronic, inflammatory autoimmune skin diseases with interface dermatitis significant morbidity whose pathogenesis is incompletely understood. Recently, NanoString Technologies has developed GeoMx® digital spatial profiling (DSP), a novel highly multiplex method for detection of proteins RNA in fixed tissues. Here, we perform proteomics transcriptomics using DSP to further characterize immune activation immunoregulatory function, including checkpoints, these overlapping diseases. We utilized 58-marker antibody cocktail found that DLE LP express unique combination inhibitory receptors. For example, programmed death receptor ligand 1 (PD-L1) lymphocyte-activation gene 3 (LAG3) detected greater amounts than LP, while expresses more T-cell immunoglobulin mucin-domain containing-3 (TIM3). next used the Cancer Transcriptome Atlas (>1800 genes) on separate cohort patients LP. Within CD3 enriched areas, there were 17 differentially expressed genes IRF7, STAT2, TNFSF10 type I IFN (IFITM1, MX1, LY6E). In contrast, IFNA17 NK cell associated KIR2DL1. IFN-g related not significantly different, suggesting both have strong II signatures, an overall stronger signature addition TNF NF-kb signaling. There few limitations our study. Our study its conclusions limited by small discovery (n=6). As such, should be taken as preliminary demonstration how can generate insights into checkpoint expression pathway DLE.

Immunohistochemistry profile of inflammatory cells in lichen planopilaris and discoid lupus erythematosus
Background: Scarring (cicatricial) alopecia represents a complex group of inflammatory disorders, mainly characterized by destruction of the hair follicle unit. Lichen planopilaris (LPP) and discoid lupus erythematosus (DLE) are the two main causes of primary cicatricial alopecia (PCA), both leading to hair follicle destruction and irreversible alopecia. However, they are different in pathogene...
متن کاملMetabolomics approach reveals urine biomarkers and pathways associated with the pathogenesis of lupus nephritis
Objective(s): lupus nephritis (LN) is a severe form of systemic lupus erythematosus (SLE) with renal complications. Current diagnosis is based on invasive renal biopsy and serum antibodies and complement levels that are not specific enough. The current study aims to identify new biomarker candidates for non-invasive diagnosis of LN and explore the pathogenic mechanisms...
متن کاملSuccessful long-term thalidomide therapy for discoid lupus erythematosus-lichen planus overlap syndrome.
Although thalidomide is a U.S. Food and Drug Admistration (FDA) approved medication for erythema nodosum leprosum and multiple myeloma, it has many off-label uses, including for discoid lupus erythematosus (DLE), Behçet's disease, apththous ulcers in HIV patients, and prurigo nodularis. Herein, we present a patient with an overlap of discoid lupus erythematosus and lichen planus who was success...
متن کاملDifferences in extracellular matrix proteins, epidermal growth and differentiation in discoid lupus erythematosus, lichen planus and the overlap syndrome.
Lichen planus (LP) and discoid lupus erythematosus (DLE) are separate disease entities. Nevertheless, patients with a so-called "overlap syndrome" have been described occasionally. The aim of the present study was to establish whether the LE/LP overlap syndrome, based on clinical and routine histological features, could be delineated from DLE or LP using immunohistochemical techniques. Formalin...

Objective: To compare the characteristics of Blaschkoian lichen planus with typical lichen planus. Patients and methods In this case series, we diagnosed 7 patients of Blaschkoian lichen planus (3 males and 4 females) based on specific clinical presentation, histopathological findings and exclusion of other dermatologic disease mimicking Blaschkoian lichen planus. Results Of 7 patients, 4 (57.1...
