β-catenin inhibition in tumor microenvironment macrophages promotes immunogenicity

Abstract Melanoma cells activate Wnt/β-catenin signaling in the tumor microenvironment (TME) to promote progression and suppress dendritic cell-mediated anti-tumor immunity. However, role of β-catenin macrophages its effect on immunity is largely undefined. In this study, using XAV939, a tankyrase inhibitor encapsulated nanoparticles, we explored whether targeting can immunogenicity TME. Our data show that co-culture with melanoma cell supernatants or presence XAV939 nanoparticles significantly promotes expression CD80 CD86, suppresses PD-L1 CD206 compared treated control nanoparticles. Similarly, nanoparticle treatment conditions resulted increased TNF-α IL-6 reduced IL-10 production by groups. suggest targeted inhibition associated could overcome tolerance
