α-Aminoadipic acid metabolism is controlled by the glutathione-dependent redox environment in Arabidopsis

Abstract α-Aminoadipic acid (Aaa) is an intermediate product in the Lys catabolism, on saccharopine pathway. Since content was increased by various abiotic stress factors affecting redox environment, a glutathione-dependent regulation of Aaa metabolism assumed which not tested earlier. The effect modified size and state glutathione pool (reduced + oxidised, GSH GSSG) related amino transcript levels compared wild-type Col-0 plants GSH-deficient pad2-1 Arabidopsis mutant with without (control) treatment. Modification resulted alteration Lys, Glu, Pro contents transcription three investigated genes Both expression two encoding enzymes its synthesis (ketoglutarate reductase/saccharopine dehydrogenase, LKR/SDH; aminoadipic semialdehyde AASADH) lower mutant, these parameters, together GSSG content, were treatment both plants. ratio exhibited positive correlation catabolic gene (dihydrolipoamide-succinyltransferase), indicates GSH-dependent control metabolism. This latter process turn can affect environment response because NADPH use/formation several steps.
